IL-37对大鼠慢性子宫内膜炎的疗效

摘　　要：目的:探讨IL-37对慢性子宫内膜炎(CE)大鼠的疗效。方法:选取10~12周龄体重220~240 g健康雌性SD大鼠30只,分为3组,每组10只。空白组不做药物处理;模型组和IL-37组均采用宫腔内推注苯酚胶浆方法建立CE模型,术后分别腹腔注射生理盐水1 mL/d或IL-37蛋白10μg/(kg·d)+生理盐水1 mL/d,连续21 d。处死大鼠,取双侧子宫,计算子宫指数,HE染色观察子宫内膜组织病理改变;免疫组化法和Western blot法检测子宫内膜组织中NLRP3、IL-1β、IL-6蛋白的表达,实时荧光定量PCR法检测3者mRNA的表达。结果:空白组、模型组和IL-37组子宫指数分别为(0.66±0.08)、(1.04±0.14)和(0.84±0.06),3组比较差异有统计学意义(P<0.001);模型组较空白组增加,IL-37组较模型组减小(P<0.001)。与空白组相比,模型组NLRP3、IL-1β、IL-6蛋白和mRNA表达水平均增加(P<0.05),IL-37组较模型组下降(P<0.05)。结论:IL-37可以明显改善CE大鼠子宫内膜炎症反应。Aim:To investigate the efficacy of IL-37 on rats with chronic endometritis(CE).Methods:A total of 30 healthy female SD rats weighing about 220-240 g at the age of 10-12 weeks were selected and divided into 3 groups,with 10 rats in each group.The blank group was not treated with medication;both the model group and the IL-37 group were intrauterine bolus injection of phenol paste to establish the CE model,and given injection of 1 mL/d of normal saline or 10μg/(kg·d)of IL-37 protein+1 mL/d of normal saline intraperitoneally for 21 days.The rats were sacrificed,bilateral uterus was taken,the uterine index was calculated,and the pathological changes of uterine tissue were observed by HE staining.The expression of proteins of NLRP3,IL-1βand IL-6 in uterine tissues was detected by immunohistochemistry and Western blot,and the mRNA expression were detected by RT-qPCR.Results:The uterine index of the blank group,the model group and the IL-37 group was(0.66±0.08),(1.04±0.14)and(0.84±0.06),respectively,and the difference among the 3 groups was significant(P<0.001);that in the model group increased compared with the blank group,and that in the IL-37 group decreased compared with the model group(P<0.001).Compared with the blank group,the protein and mRNA expression levels of NLRP3,IL-1βand IL-6 in the model group increased(P<0.05),while those of the IL-37 group decreased compared with the model group(P<0.05).Conclusion:IL-37 can significantly improve the intimal inflammatory response in CE rats.
