摘 要:目的:探究更有效的脱细胞方法,用于制备组织工程小口径血管。方法:通过两种不同的脱细胞方法,制备小口径脱细胞血管支架,并测量组织及理化性质。方法:将新鲜绵羊颈内动脉分为A组:对照组、B组:酶法脱细胞组(实验组Ⅰ)、C组:酶与去污剂混合脱细胞组(实验组Ⅱ)。将三组进行HE染色、弹力纤维染色、胶原纤维染色、DAPI荧光染色、电镜观察。测量理化性质如厚度、拉力强度、含水量、DNA含量、承受压力值。结果:B组厚度最小;C组拉力强度最大;C组含水量和DNA含量最少;三组均可承受300mm Hg(1 mmHg=0.133kPa)压力值。结论:两种脱细胞方法均可去除血管壁细胞并损伤血管固有组织。C组脱细胞程度最高,DNA含量最少,对血管固有组织损伤最小。Objective:To find the better method to decellularization for tissue engineered small-diameter vessels.Methods:We contrasted with two different methods to decellularize sheep internal carotid arteries as tissue engineered small-diameter vascular scaffold and monitored histopathology,immunofluorescence,biomechanics.Blood vessels were divided into three groups.Group A:fresh group(control group),Group B:enzymatic decellularization group(experimental groupⅠ),Group C:the enzyme-detergent mixing decellularization group(experimental groupⅡ).The experimental groups were treated with two decellularization methods respectively.HE staining,elastic and collagenic fiber dyeing,fluorescence staining,electronic microscopy were observed,and the thickness,tensile strength,moisture content,DNA content,burst pressure had been tested,recorded and analyzed.Results:Group B has least thickness.Group C has most tensile strength,least moisture and DNA contents.All of three groups could endure pressure more than 300mmHg.Conclusions:Both methods could remove cells and damage vessel walls.The enzyme-detergent mixing decellularization method demonstrates higher level of decellularization,less DNA residual and milder to damage vessels.
