重庆某“区域长寿”聚集区不同年龄段老年男性的mtDNA变异率分析

摘  要:目的研究重庆某“区域长寿”聚集区不同年龄段老年男性的生活习惯和血生化指标的差异及高龄男性老年人(≥90岁)线粒体DNA(mtDNA)的高频变异情况。方法随机选择重庆某“区域长寿”聚集区≥60岁老年男性作为研究对象,依据年龄分为A组(60~<70岁)、B组(70~<80岁)、C组(80~<90岁)、D组(≥90岁,高龄),比较不同年龄组间蔬菜水果食用情况、奶类豆类食用情况、户外活动情况、睡眠时间、血生化指标的差异;然后对上述人员采集外周血提取mtDNA并测序,通过生物信息学分析找出各组与D组间差异有统计学意义的mtDNA突变。结果不同年龄组间蔬菜水果食用情况、奶类豆类食用情况、户外活动情况、睡眠时间比较差异无统计学意义(P>0.05);血生化指标中红细胞和肌酐在部分组间差异有统计学意义(P<0.05);线粒体mtDNA突变分析共发现4105个突变位点,以单碱基突变为主,最常见的突变为T>C(22.57%),其次是C>T(19.82%)、A>G(19.36%);D组mtDNA平均突变率最高,为8.392%,其中共发现3个mtDNA变异在不同年龄组与D组比较间差异均有统计学意义(P<0.1),包括:mtDNA5396-5411CAACGTAAA>C、mtDNA5414A>T、mtDNA5417G>T。结论不同年龄段男性老年人生活习惯无明显差异;高龄组mtDNA突变率最高;mtDNA5396-5411CAACGTAAA>C、mtDNA5414A>T、mtDNA5417G>T变异可能与“区域长寿”相关。Objective To study the differences in living habit and blood biochemical indicators of male elderly people at different age groups in a“regional longevity”gathering area in Chongqing and the high frequency variation of mitochondrial DNA(mtDNA)in the elderly people(≥90 years old).Methods The male elderly people aged≥60 years old in a“regional longevity”gathering area in Chongqing were randomly selected as the study subjects and divided into the group A(60-<70 years old),group B(70-<80 years old),group C(80-<90 years old)and group D(≥90 years old).The differences of vegetable and fruit consumption,dairy bean consumption,outdoor activities,sleep time and blood biochemical indexes were compared among different age groups.Then peripheral blood in the above subjects was collected for extracting mtDNA and sequencing.The meaningful mtDNA mutations between the various groups and group D were found out by the bioinformatics analysis.Results There were no statistically significant differences in consumption of vegetables and fruits,consumption of dairy beans,outdoor activities and sleep duration among different age groups(P>0.05).There were statistically significant differences in erythrocyte and creatinine of blood biochemical indexes among the partial groups(P<0.05).The mitochondria mutation analysis found 4105 mtDNA mutation sites,mainly single base mutation,the most common mutation was T>C(22.57%),followed by C>T(19.82%)and A>G(19.36%).The average mutation rate of mtDNA in the group D was the highest(8.392%),in which 3 mtDNA variants were found to have significant differences between different age groups and group D(P<0.1),including mtDNA5396-5411CAACGTAAA>C,mtDNA5414A>T and mtDNA5417G>T.Conclusion There was no significant difference in the living habits of male elderly people of different ages.The mutation rate of mtDNA in the advanced age group was the highest.mtDNA5396-5411CAACGTAAA>C,mtDNA5414A>T and mtDNA5417G>T variations may be related to“regional longevity”.
