摘 要:目的:探究槐角苷对去势大鼠骨质疏松的作用及机制。方法:80只雌性SD大鼠随机分为假手术组10只和去势组70只,70只去势大鼠随机分为模型组、骨松宝颗粒组、阿仑膦酸钠组、雌激素组、槐角苷高、中、低剂量组,每组10只,给药治疗6个月,分离大鼠血清和股骨组织。ELISA法检测大鼠血清骨特异性碱性磷酸酶(BAP)、抗酒石酸酸性磷酸酶(TRAP)、高密度脂蛋白(HDL-C)、低密度脂蛋白(LDL-C)、三酰甘油(TG)水平,HE染色和masson染色观察股骨组织的病理变化,免疫组化法检测股骨组织β-连环蛋白(β-catenin)、核因子κB受体活化因子配体(RANKL)的阳性表达,RT-PCR检测股骨组织RANKL、β-catenin mRNA表达。结果:模型组大鼠的骨代谢指标BAP、TRAP较于假手术组明显升高,脂代谢指标LDL-C、TG表达升高,HDL-C表达降低;骨小梁厚度变薄,骨髓腔变大,软骨基质无明显钙化;骨组织β-catenin蛋白及mRNA表达显著降低(P<0.01),RANKL蛋白及mRNA表达显著升高(P<0.01)。与模型组比较,槐角苷高剂量组骨代谢及脂代谢指标有所改善;骨小梁厚度增厚,髓腔变小,软骨基质有所钙化;β-catenin蛋白及mRNA表达显著增加(P<0.01),RANKL蛋白及mRNA显著降低(P<0.01)。结论:槐角苷对去势大鼠具有抗骨质疏松作用,其作用机制与激活Wnt/β-catenin信号通路,刺激成骨细胞生长和调控RANK/RANKL信号通路抑制破骨细胞分化有关。Objective:To investigate the effect and mechanism of Sophoricoside on osteoporosis in castrated rats.Methods:80 female SD rats were randomly divided into sham operation group(10)and castrated group(70).70 castrated rats were randomly divided into model group,Gusongbao granule group,alendronate sodium group,estrogen group and Sophoricoside high-dose,medium-dose and low-dose groups(10 in each group).The serum and femoral tissue of rats were separated after 6 months of treatment.Serum levels of bone-specific alkaline phosphatase(BAP),tartrate-resistant acid phos-phatase(TRAP),high density lipoprotein(HDL-C),low density lipoprotein(LDL-C)and triacylglycerol(TG)were detected by ELISA.HE staining and Masson staining were used to observe the pathological changes of femur tissue,the positive expressions of B-catenin and nuclear factor KB receptor activating factor ligand(RANKL)in femur tissue were detected by immunohistochemistry,and the mRNA expressions of RANKL and B-catenin in femur tissue were detected by RT-PCR.Results:Bone metabolism indexes BAP and TRAP in model group were significantly higher than those in sham group,lipid metabolism indexes LDL-C and TG were increased,and HDL-C expression was decreased.Bone trabecular thickness was thinner,bone marrow cavity was larger,and cartilage matrix was not calcified.The expression of B-catenin protein and mRNA in bone ti-ssue was significantly decreased(P<0.01),and the expression of RANKL protein and mRNA was significantly increased(P<0.01).Compared with model group,bone metabolism and lipid metabolism indexes in Sophorico-side high dose group were improved.The thickness of bone trabeculae increased,the pulp cavity became sma-ller,and the cartilage matrix calcified.The expression of B-catenin protein and mRNA increased significantly(P<0.01),and RANKL protein and mRNA decreased significantly(P<0.01).Conclusion:Sophoricoside has anti-osteoporosis effect on castrated rats,and its mechanism is related to activating Wnt/B-catenin signa-ling pathway to stimulate osteoblast gro
