摘 要:为了研究枯草芽孢杆菌菌液OD值与活菌数之间的相互关系,建立一种能快速获得菌悬液活菌数量的方法。利用分光光度法分别测定不同稀释度菌液在500、550、580、600、630、650、680 nm处的OD值,确定其最适吸收波长。再结合稀释涂布平板法测得各菌液的活菌数,比较二者之间的相关性,建立线性回归方程。结果表明,600 nm为枯草芽孢杆菌的最适吸收波长,其活菌数量与OD600呈直线线性相关,其回归方程为:y=1.4935x-0.0276,相关系数R2=0.9922。证明在一定的浓度范围内采用分光光度法测定枯草芽孢杆菌活菌数的方法是可行的,且更简便快捷。In order to study the relationship between the OD value of Bacillus subtilis bacterial solution and the number of viable bacteria,a method to quickly obtain the number of viable bacteria in bacterial suspension was established.Measure the OD values of bacterial solutions with different dilutions at 500,550,580,600,630,650,and 680 nm using spectrophotometry to determine the optimum absorption wavelength.The number of viable bacteria in each bacterial solution was measured by the dilution coating plate method,and the correlation between them was compared,and the linear regression equation was established.The results showed that 600 nm was the optimal absorption wavelength for Bacillus subtilis,and the number of viable bacteria was linearly correlated with OD600.The regression equation was y=1.4935x-0.0276,and the correlation coefficient R^(2)=0.9922.It is proved that the method of determining the number of Bacillus subtilis viable by spectrophotometry within a certain concentration range is feasible and more convenient and efficient.
