摘 要:【目的】研究仿刺参(Apostichopus japonicus)体腔液的细菌凝集活性,为寻找仿刺参病害生态学防控方法提供依据。【方法】采用玻片法分别考察仿刺参体腔液上清和体腔细胞破碎液上清对灿烂弧菌(Vibriosplendidus)、哈维弧菌(Vibrio harveyi)、鳗弧菌(Vibrio anguillarum)、溶藻弧菌(Vibrio alginolyticus)、金黄色葡萄球菌(Staphylococcus aureus)和大肠埃希菌K12(Escherichia coli K12)的凝集活性,同时研究α-D-葡萄糖、D-半乳糖、D-木糖、D-山梨醇、蔗糖、麦芽糖、α-D-乳糖、甘露醇和N-乙酰神经氨酸等9种糖和Ca^(2+)、Zn^(2+)、Mn^(2+)、Mg^(2+)、Cd^(2+)、Fe^(2+)等6种二价金属离子对仿刺参体腔液细菌凝集活性的影响。【结果】仿刺参体腔液上清和体腔细胞破碎液上清对6种受试菌均有凝集作用,前者凝集比活为14.0~35.0μg/mL,对鳗弧菌和大肠埃希菌K12的凝集作用最大,对溶藻弧菌的凝集作用最小,后者凝集比活为6.0~7.5μg/mL,对灿烂弧菌的凝集作用最大,对其余5种受试菌的凝集作用相同。此外,α-D-葡萄糖可全部抑制体腔液对6种受试菌的凝集活性,麦芽糖、N-乙酰神经氨酸、D-山梨醇和甘露醇能部分抑制其细菌凝集活性,最低凝集抑制浓度范围为100~200 mmol/L,α-D-乳糖、D-半乳糖、D-木糖和蔗糖对其细菌凝集活性无明显影响;Ca^(2+)、Zn^(2+)、Mn^(2+)可促进仿刺参体腔液上清的细菌凝集活性,6种二价金属离子对体腔细胞破碎液上清的细菌凝集活性无明显影响。【结论】仿刺参体腔液上清和体腔细胞破碎液上清均具有细菌凝集活性,其凝集活性能被α-D-葡萄糖全部抑制,被N-乙酰神经氨酸、甘露醇、D-山梨醇和麦芽糖部分抑制,且Ca^(2+)、Zn^(2+)、Mn^(2+)可促进仿刺参体腔液上清的细菌凝集活性。【Objective】This study aims to investigate the bacterial agglutination activity of Apostichopus japonicus,providing a foundational basis for the ecological prevention and control of this species.【Method】This study used the slide test to determine the agglutination activity of the supernatant of coelomic fluid and coelomocyte lysate against Vibrio splendidus,Vibrio harveyi,Vibrio anguillarum,Vibrio alginolyticus,Staphylococcus aureus and Escherichia coli K12,and then determined the effects of 9 kinds of saccharides(includingα-D-glucose,D-galactose,D-Xylose,D-sorbitol,sucrose,maltose,α-D-lactose,mannitol and N-acetylneuraminic acid)and 6 kinds of divalent metal ions(including Ca^(2+),Zn^(2+),Mn^(2+),Mg^(2+),Cd^(2+)and Fe^(2+))on the bacterial agglutinative activity of the coelomic fluid in A.japonicus.【Results】The results showed that both the supernatant of coelomic fluid and coelomocyte lysate had agglutinative effects on the six tested bacteria.Among them,the agglutination activity of coelomic fluid supernatant is 14.0 to 35.0μg/mL,with highest agglutination effect on V.anguillarum and E.coli K12,while showing the lowest level of agglutination with V.alginolyticus.The agglutination activity of coelomocyte lysate supernatant is 6.0 to 7.5μg/mL,which has the greatest agglutination effect on V.splendidus and has the same bacterial agglutinative activity on the other five bacteria.Additionally,α-D-glucose could completely inhibit the agglutination activity of six bacteria,while maltose,N-acetylneuraminic acid,D-sorbitol and mannitol could partially inhibit the agglutination activity of these bacteria,with a minimum inhibitory concentration of 100-200 mmol/L.α-D-lactose,D-galactose,D-Xylose and sucrose had no significant effect on the bacterial agglutinative activity.In addition,Ca^(2+),Zn^(2+)and Mn^(2+)increased the agglutinative activity of the coelomic fluid supernatant in A.japonicus,while 6 kinds of divalent metal ions showed no obvious effect on the agglutinative activity of coelomocyte lysate
