基于离线二维色谱-质谱联用技术的何首乌化学成分表征研究

摘  要:目的基于液质联用技术对何首乌九蒸九晒炮制品的化学成分进行表征。方法通过优化离线二维色谱的固定相、流动相、柱温、进样量等条件,构建了离线二维液相色谱与四极杆-静电场轨道阱高分辨质谱联用的方法(2D-LC-Q-Orbitrap MS),表征何首乌炮制品中的化学成分,并对其裂解规律进行解析。结果构建了由亲水相互作用色谱柱XBridge Amide(4.6 mm×150 mm,3.5μm)与反相色谱柱CORTECS Shield RP18(2.1 mm×100 mm,1.7μm)组成的离线二维液相色谱系统,通过对何首乌炮制品中的化学成分进行定性分析,共表征了139个化学成分。结论本文构建的离线二维液相色谱相比于一维色谱更有利于何首乌化学成分的表征,可为探寻何首乌中的药效和毒性物质基础提供数据支持,为后续药理、毒理研究提供参考。Objective To characterize the chemical constituents of Polygonum multiflorum Thunb.after steaming and drying times.Methods By optimizing the off-line two-dimensional stationary phase,mobile phase,column temperature,sample volume and other conditions,we established offline two-dimensional liquid chromatography and quadrupole-electrostatic field orbitrap high-resolution mass spectrometry(2D-LC-Q-Orbitrap MS)to analyze the cracking law of chemical constituents in Polygonum multiflorum Thunb..Results An off-line two-dimensional liquid chromatography system consisting of hydrophilic interaction chromatography XBridge Amide(4.6 mm×150 mm,3.5μm)and reversed-phase chromatography CORTECS Shield RP18(2.1 mm×100 mm,1.7μm)was established.Totally 139 chemical constituents were characterized by qualitative analysis of the constituents in the processed Polygonum multiflorum Thunb..Conclusion This study provides a reference for exploring the material basis of liver injury caused by Polygonum multiflorum Thunb.,and for subsequent pharmacological and toxicological research.
