摘  要:目的考察草归除湿活血颗粒剂的提取工艺,并探究其通过降尿酸治疗高尿酸症的机制。方法通过HPLC测定草归除湿活血颗粒水提液中各成分的含量,筛选最佳提取条件;利用UPLC-Q-TOF-MS分析其活性成分,再将得到的活性成分结合高尿酸血症的潜在靶点,进行PPI网络构建及GO、KEGG分析。结果最佳提取工艺为10倍水浸泡0.5 h,加热回流提取2次,每次1 h。UPLC-Q-TOF-MS得221个活性成分,网络药理学分析得到183个潜在交集靶点,富集分析得到PI3K-Akt信号通路、脂质和动脉粥样硬化、HIF-1等信号通路。结论草归除湿活血颗粒可能通过调节PI3K-Akt信号通路、脂质和动脉粥样硬化、HIF-1等信号通路来发挥降尿酸的作用。Objective To determine the extraction process of Caogui Chushi Huoxue granules,and its mechanism in lowering the uric acid for hyperuricemia.Methods The content of the components in the aqueous extract of Caogui Chushi Huoxue granules was determined by HPLC,and the optimal extraction conditions were obtained.The active components were analyzed by UPLC-Q-TOF-MS,and then the obtained active components were combined with hyperuricemia targets to serve as potential targets.PPI network was constructed and GO and KEGG analyses conducted.Results The optimal extraction process included 10-fold water soaked for 0.5 h,heating,and reflux extraction twice,1 h for each.Totally 221 active components were obtained by UPLC-Q-TOF-MS,and 183 potential targets by PPI network.The signaling pathways,such as PI3K-Akt pathway,lipid and atherosclerosis,and HIF-1,were obtained by enrichment analysis.Conclusion Caogui Chushi Huoxue granules may regulate the signaling pathways such as TNF,PI3K-Akt,and cancer to lower the uric acid.
