龙藤复方对类风湿关节炎大鼠模型的治疗作用研究

摘　　要：目的观察龙藤复方对佐剂型关节炎(AA)大鼠足跖肿胀度、关节炎指数(AI)、血清肿瘤坏死因子(TNF)-α、白细胞介素(IL)-1β、IL-6水平和踝关节病理学的影响。方法66只SD大鼠随机分为正常组,模型组,阳性药雷公藤多苷片组(9 mg·kg^(-1)),龙藤复方低剂量组(0.435 g·kg^(-1)),龙藤复方中剂量组(0.87 g·kg^(-1))及龙藤复方高剂量组(1.74 g·kg^(-1))。于大鼠左后足跖皮内注射弗氏完全佐剂(CFA)致炎建立AA模型,致炎后第15日各给药组按剂量灌胃给药。于实验d1、d7、d14、d21、d28对各组大鼠进行AI评分,计算足跖肿胀度。实验d28处死大鼠,腹主动脉取血,离心后得到血清,酶联免疫吸附法(ELISA)测定血清中TNF-α、IL-1β、IL-6水平,取踝关节HE染色观察病理变化。结果给药14 d(造模28 d)后,与模型组大鼠比较,龙藤复方各剂量组及雷公藤多苷组AI评分,足跖肿胀度,血清TNF-α、IL-6、IL-1β含量明显降低(P<0.05或0.01);龙藤复方中、低剂量组和阳性药组均明显改善踝关节病理变化。结论龙藤复方对AA大鼠具有治疗作用,作用机制与其降低血清中TNF-α、IL-1β、IL-6水平,改善踝关节病理损伤有关。Objective To determine the effect of Longteng compound on foot-plantar swelling,arthritis index,serum tumor necrosis factor(TNF)-α,interleukins(IL)-1β,IL-6 levels and ankle joint pathology in rats with adjuvant arthritis.Methods Totally 66 SD rats were randomly divided into a normal group,a model group,a positive drug tripterygium glycosides group(9 mg·kg^(-1)),a lowdose Longteng compound group(0.435 g·kg^(-1)),a medium-dose Longteng compound group(0.87 g·kg^(-1)),and a high-dose Longteng compound group(1.74 g·kg^(-1)).Inflammation was induced by intradermal injection of Freund’s complete adjuvant(CFA)into the left hindfoot of the rats,resulting in an adjuvant arthritis model,and the drug was administered by gavage to each group at the corresponding dose on the 15th day of the inflammation period.The arthritis index was scored on d1,d7,d14,d21,and d28,and the swelling of the plantar foot was measured.The rats were sacrificed on d28.Blood was collected from the abdominal aorta and centrifuged to obtain the serum.The levels of TNF-α,IL-1β,and IL-6 in the serum were determined by enzyme-linked immunosorbent assay,and the ankle joints were taken to observe the pathological changes by HE staining.Results After 14 d of drug administration(28 d of modeling),the index scores,foot-plantar swelling,and the serum TNF-α,IL-6,and IL-1βlevels were significantly reduced in all dose groups of Longteng compound and tripterygium glycosides when compared with those of the model group(P<0.05 or 0.01).The ankle joint pathology was improved in the medium-dose,and low-dose Longteng compound groups and the positive group.Conclusion Longteng compound has therapeutic effect on rats with adjuvant arthritis,and the mechanism may be related to its reduction of TNF-α,IL-1βand IL-6 levels in the serum and improvement in the pathological damage of ankle joints.
