防风炒炭前后HPLC指纹图谱和4种色原酮成分含量变化研究

摘  要:目的建立防风及防风炭HPLC指纹图谱,考察不同产地防风炒炭前后4种色原酮类成分含量,探究防风炒炭过程中各成分变化规律。方法采用Venusil C18 Plus(4.6 mm×250 mm,5μm),以水-甲醇梯度洗脱,流速1.0 mL·min^(-1),检测波长254 nm,柱温30℃。建立防风及防风炭HPLC指纹图谱,通过统计学方法对防风炒炭前后指纹图谱进行分析,并测定升麻素苷、5-O-甲基维斯阿米醇苷、升麻素和亥茅酚苷的含量。结果不同产地防风及防风炭HPLC指纹图谱存在差异。各产地防风炒炭后升麻素苷及5-O-甲基维斯阿米醇苷含量均降低,平均降幅分别为17.94%,6.15%;其中升麻素降低显著,平均降幅为21.14%;亥茅酚苷含量呈上升趋势,平均增幅为10.65%。结论防风炮制前后指纹图谱存在差异,4种色原酮类成分含量变化,可能与防风炭偏于止血有关。本研究为防风炭中活性物质及质量控制提供参考。Objective To establish the HPLC fingerprints of Saposhnikovia divaricata and Saposhnikovia divaricata charcoal,and determine the content of 4 chromones in Saposhnikovia divaricata charcoal from different origins before and after the charcoal-frying,as well as the changing patterns of various components in the process of charcoal-frying.Methods Venusil C18 Plus(4.6 mm×250 mm,5μm)was used with water-methanol gradient elution at a flow rate of 1.0 mL·min^(-1),a detection wave length of 254 nm,and a column temperature of 30℃.HPLC fingerprints of Saposhnikovia divaricata and Saposhnikovia divaricata charcoal were established,and the fingerprints of Saposhnikovia divaricata before and after the charcoal-frying were analyzed statistically.The content of prim-O-glucosylcimifugin,4'-O-β-D-glucosyl-5-O-methylvisamminol,cimifugin,and sec-O-glucosylhamaudol were determined.Results There were differences in the HPLC fingerprints of Saposhnikovia divaricata and Saposhnikovia divaricata charcoal from different origins.The content of prim-O-glucosylcimifugin and 4'-O-β-D-glucosyl-5-O-methylvisamminol in charcoal-fried Saposhnikovia divaricata from various origins decreased by 17.94%and 6.15%on average.Cimifugin decreased most,with an average decrease of 21.14%.The content of sec-O-glucosylhamaudol increased,with an average growth of 10.65%.Conclusion There are differences in the fingerprints of Saposhnikovia divaricata charcoal before and after the processing.The content of 4 chromones varies,which may be related to the hemostatic effect of Saposhnikovia divaricata charcoal.This study provides a reference for the study of active substances and quality control of Saposhnikovia divaricata charcoal.
