摘 要:目的:探讨应用全长16S-rRNA基因测序技术指导盆腔器官脱垂患者应用引导菌群调节药物治疗的重要性。方法:选取2022年1月—2024年6月三明市第一医院接受子宫托治疗的80例盆腔器官脱垂患者(观察组)以及80名健康体检者(对照组),对比分析两组阴道微生态状况及16S-rRNA测序结果;基于检测结果,对观察组中阴道微生态失衡的患者实施乳酸杆菌栓剂治疗,进行二次分组,即观察1组(未接受药物干预)、观察2组(接受乳酸杆菌栓剂治疗),比较两组阴道微生态和16S-rRNA测序结果。结果:观察组与对照组在阴道菌群密度及优势菌群构成上存在差异(P<0.05),且16S-rRNA测序结果显示观察组中乳酸杆菌等厚壁菌门数量较少,而加德纳菌等放线菌门数量较多。观察2组经过干预治疗,该组阴道微生态失衡状况得到改善,厚壁菌门数量显著增多,而观察1组阴道微生态则面临失衡风险。结论:在盆腔器官脱垂患者中,16S-rRNA测序技术能够有效分析阴道菌群变化,并为阴道菌群调节药物的使用提供依据,为促进阴道微生态平衡、巩固脱垂治疗效果提供新的策略和方法。Objective:To explore the importance of using full-length 16S-rRNA gene sequencing technology to guide the application of guided flora to regulate drug therapy in patients with pelvic organ prolapse.Methods:A total of 80 patients with pelvic organ prolapse(observation group)and 80 healthy subjects(control group)who received pessary treatment in Sanming First Hospital from January 2022 to June 2024 were selected.The vaginal microecology and 16S-rRNA sequencing results of the two groups were compared and analyzed.Based on the test results,the patients with vaginal microecological imbalance in the observation group were treated with lactobacillus suppository,and the second group was divided into observation group 1(without drug intervention)and observation group 2(receiving lactobacillus suppository treatment).The vaginal microecology and 16 S-rRNA sequencing results of the two groups were compared.Results:There were differences in vaginal flora density and dominant flora composition between the observation group and the control group(P<0.05),and 16S-rRNA sequencing results showed that the number of Firmicutes such as Lactobacillus in the observation group was less,while the number of Actinobacteria such as Gardnerella was more.After intervention treatment,the imbalance of vaginal microecology in the observation group 2 was improved,and the number of Firmicutes was significantly increased,while the vaginal microecology in the observation group 1 was at risk of imbalance.Conclusion:For patients with pelvic organ prolapse,16S-r RNA sequencing technology can effectively analyze the changes of vaginal flora,provide a basis for the use of vaginal flora regulating drugs,and provide new strategies and methods for promoting vaginal microecological balance and consolidating the therapeutic effect of prolapse.
