摘  要:目的 确定骨结核(bone tuberculosis,BT)患者肠道微生物的差异性生物标志物。方法 采用扩增测序法对BT患者的肠道菌群特征进行分析。纳入30例健康者(HC组)和30例BT患者(BT组)为研究对象,收集各组对象粪便样本并提取基因组DNA进行16S rDNA V3-V4区测序。评价比较肠道菌群alpha和beta多样性以及肠道细菌谱,并采用随机森林算法构建基于肠道菌群的BT预测诊断模型。结果 与HC组相比,BT组患者肠道菌群存在显著差异(P<0.05)。BT组患者的肠道菌群以alpha多样性降低为特征,同时BT组患者肠道乳杆菌科、气球菌科、埃希-志贺菌属等菌群丰度较高。两组人群之间存在51条差异代谢通路。结论 BT患者肠道菌群出现特异性改变,肠道菌群中BT相关微生物的鉴定可能有助于我们寻求更多的临床治疗方法。Objective To identify the differential biomarkers of gut microbiota in patients with bone tuberculosis(BT).Methods The amplification sequencing method was used to characterize the features of gut microbiota in BT patients.16S rDNA V3−V4 region sequencing was performed on fecal DNA isolated from 30 healthy controls(HC group)and 30 BT patients(BT group).The alpha and beta diversities,and gut microbiome profiles of these two groups were processed and assessed.A gut microbiota-based model for the predictive diagnosis of BT was constructed using a random forest algorithm.Results Compared with HC group,there were significant differences in microbial communities in BT group(all P<0.05).The gut microbiome of BT patients was characterized by the decreased alpha diversity,with a higher content of Lactobacillaceae,Aerococcaceae and Escherichia_Shigella,etc.Besides,there were 51 different metabolic pathways between these two groups.Conclusion There are specific alterations in the gut microbiota in BT patients.Identification of BT-related microorganisms in the gut microbiota may help us seek more treatment methods.
