摘  要:【目的】建立一种实时、动态检测外源酶催化烟叶水解反应的方法。【方法】利用微量热仪研究果胶酶催化烟叶水解反应的热动力学特性。【结果】果胶酶催化烟叶水解反应可分为两个阶段,I阶段是酶液与烟叶接触后,热流呈直线上升,达到峰值,II阶段是热流从峰值开始缓慢平稳下降,逐渐回到基线水平;通过热流-时间曲线,可以获得反应时间T、峰值P_(max)、放热量Q、反应速率常数k和反应进度φ等水解反应过程的热动力学参数;依据热流曲线及参数,能够较好地表征烟叶与酶制剂水解反应的差异。【结论】微量热法能够实时、动态、定性、定量表征果胶酶催化烟叶水解反应过程。[Objective]This study aims to establish a real-time and dynamic method for detecting the hydrolysis of tobacco catalyzed by exogenous enzymes.[Methods]The thermodynamic characteristics of pectinase-catalyzed hydrolysis of tobacco were studied by microcalorimeter.[Results]The results indicate that the pectinase-catalyzed hydrolysis of tobacco can be divided into two stages:Stage I,where the heat flow rises linearly to a peak after the enzyme solution contacts the tobacco leaves;and Stage II,where the heat flow gradually and steadily declines from the peak back to the baseline level.By analyzing the power-time curve,various thermodynamic parameters of the hydrolysis process can be obtained,including reaction time T,peak heat value P_(max),heat release Q,reaction rate constant k,and reaction progress.Furthermore,the differences in the hydrolysis reaction between tobacco leaves and enzyme preparations can be well characterized based on the heat flow curve and parameters.[Conclusion]The experimental results demonstrate that microcalorimetry can real-time,dynamically,qualitatively,and quantitatively characterize the hydrolysis reaction process of tobacco leaves catalyzed by pectinase.
