摘 要:【目的】获得对烟草棒孢霉叶斑病有防控效果的微生物菌种。【方法】利用稀释涂布平板法分离纯化烟株内生细菌,筛选出对多主棒孢霉具有较好拮抗活性的内生细菌,结合形态学和分子生物学方法开展拮抗菌株鉴定;采用浸渍法测定拮抗菌对多主棒孢霉菌丝形态、分生孢子萌发和抗氧化酶活性的影响;通过盆栽法评价拮抗菌对烟草棒孢霉叶斑病的防控效果。【结果】(1)从健康烟株根、茎及叶分离获得55株内生细菌,其中TKY-11对多主棒孢霉拮抗效果较好,抑菌率65.00%,鉴定为贝莱斯芽孢杆菌属Bacillus velezensis。(2)TKY-11菌株发酵上清液显著抑制多主棒孢霉分生孢子萌发,处理18 h时孢子萌发率仅50.67%;发酵上清液能降低多主棒孢霉菌丝的抗氧化酶活性,并破坏菌丝细胞结构。(3)接种TKY-11可有效降低烟草棒孢霉叶斑病的发病程度。【结论】本研究分离的贝莱斯芽孢杆菌TKY-11对多主棒孢霉有拮抗作用,具有一定的开发潜力。[Objective]This study aims to obtain microbial strains with control effects against Corynespora cassiicola.[Methods]Tobacco endophytic bacteria were isolated using the dilution spread plate method,and antagonistic bacteria against C.cassiicola were then screened using plate confrontation method.On this basis,morphological characterization and molecular biological methods were carried out to identify antagonistic strain.the effects of antagonistic bacteria on mycelium morphology,conidial germination,and antioxidant enzyme activities of C.cassiicola were determined using maceration method.Finally,the control effect of antagonistic strain against tobacco Corynespora leaf spot was evaluated through pot experiments.[Results](1)A total of 55 endophytic bacteria were isolated from the roots,stems,and leaves of healthy tobacco plants,among which strain TKY-11 showed good antagonistic effects against C.cassiicola with an inhibition rate of 65.00%.The strain was identified as Bacillus velezensis.(2)The fermentation supernatant of strain TKY-11 significantly inhibited the germination of C.cassiicola conidia,with a spore germination rate of only 50.67%after 18 hours of treatment.The fermentation supernatant reduced the antioxidant enzyme activity of C.cassiicola mycelium and disrupted the structure of mycelial cells.(3)Inoculation with TKY-11 effectively reduced the severity of tobacco Corynespora leaf spot.[Conclusion]The Bacillus velezensis strain TKY-11 isolated in this study exhibits antagonistic activity against C.cassiicola and shows potential for development.
