摘 要:为有效防治世界性检疫害虫马铃薯块茎蛾Phthorimaea operculella,测定不同浓度α-茄碱胁迫下马铃薯块茎蛾生长发育、解毒酶活性和细胞色素P450(cytochrome P450,CYP450)基因表达量的变化,选择相对表达量差异较显著的2个CYP450基因进行RNA干扰试验,测定干扰这2个CYP450基因后,马铃薯块茎蛾存活率、体重和龄期的变化。结果显示:α-茄碱可以抑制马铃薯块茎蛾幼虫的生长发育;α-茄碱能够诱导马铃薯块茎蛾幼虫体内CYP450酶和谷胱甘肽S-转移酶活性上升,同时抑制乙酰胆碱酯酶与羧酸酯酶活性。在α-茄碱胁迫下,马铃薯块茎蛾体内6个CYP450基因在肠道中高表达,且随着α-茄碱浓度升高,其表达量总体呈现先上升后下降的趋势。干扰CYP340CQ1和CYP6AE221基因后用α-茄碱饲喂马铃薯块茎蛾,其存活率、幼虫体重和3~4龄幼虫期均较对照发生变化。表明CYP450基因在马铃薯块茎蛾对α-茄碱的代谢解毒中可能发挥着重要作用。To effectively manage and control worldwide quarantine pest potato tuber moth Phthori-maea operculella,the effects ofα-solanine at different concentrations on the growth,development,de-toxification enzyme activity,and cytochrome P450(CYP450)gene expression of P.operculella were in-vestigated.Two CYP450 genes with significant relative expression changes were selected for RNA in-terference(RNAi)experiments.Changes in survival rate,body weight,and developmental stage of P operculella were analyzed after interference with these two CYP450 genes.The results showed thatα-solanine inhibited the growth and development of P operculella larvae.α-solanine induced an increase in the activity of CYP450 enzymes and glutathione S-transferase,while it inhibited the activity of acetyl-cholinesterase and carboxylesterase.Under aα-solanine stress,six CYP450 genes were highly expressed in larval intestines,with expression levels generally increasing initially and then declining as a-solanine concentration increased.RNAi targeting of CYP340CQ1 and CYP6AE221,followed byα-solanine feed-ing,resulted in significant changes in survival rate,larval weight,and 3rd to 4th instar larval stage of P.operculella.These findings indicate that CYP450 genes play an essential role in the detoxification and metabolism of a-solanine in P operculella.This study provides insights into the detoxification mecha-nisms ofα-solanine and offers new strategies for the prevention and control of P.operculella.
