摘 要:为探究乌拉尔甘草共生根瘤菌的微进化历程,温室条件下种植乌拉尔甘草捕捉当地土壤中根瘤菌,共分离得到根瘤菌21株,基于核心基因recA序列鉴定菌株的种属分别为豌豆根瘤菌(Rhizobium leguminosarum)、阿拉米根瘤菌(Rhizobium alamii)、阳陵根瘤菌(Rhizobium yanglingense)、菜豆根瘤菌(Rhizobium phaseoli)、圆明园慢生根瘤菌(Bradyrhizobium yuanming ense)、Rhizobium sp.I和Bradyrhizobium sp.II,其中Rhizobium sp.I为第一优势菌群,Bradyrhizobium sp.II为第二优势菌群,群落结构相对单一,核苷酸多态性(π)较低(范围为0.00205~0.01791,平均值为0.010406),遗传距离(Dxy)较远(范围为0.0688~0.1935,平均值为0.1483),基因交流指数(Nm)较低(范围为0.02~17.10,平均值为3.048)。综上所述,内蒙古东部地区乌拉尔甘草根瘤菌遗传多样性较高,在进化历史上基因交流频率较低,具有较强的遗传稳定性。To investigate the microevolution of symbiotic rhizobia associated with Glycyrrhiza uralensis,the plant was cultivated aseptically in greenhouse to isolate rhizobia from the local soil.A total of 21 rhizobial strains were isolated and identified as Rhizobium leguminosarum,Rhizobium alamii,Rhizobium yanglingense,Rhizobium phaseoli,Bradyrhizobium yuanmingense,Rhizobium sp.I and Bradyrhizobium sp.II.And Rhizobium sp.I was the predominant group,followed by Bradyrhizobium sp.II based on the core gene recA sequences.The community structure was relatively simple,with low nucleotide diversity(π)observed(range:0.00205~0.01791,mean:0.010406).Genetic differentiation(Dxy)was high,with values ranging from 0.0688 to 0.1935(mean:0.1483).The gene flow index(Nm)exhibited low values(range:0.02~17.10,mean:3.048).In conclusion,the high genetic diversity and low frequency of gene exchange suggest that the rhizobia associated with G.uralensis in eastern Inner Mongolia exhibit strong genetic stability throughout their evolutionary history.
