摘  要:目的探讨食欲肽对缺血性脑卒中(CIS)模型大鼠的神经保护作用及其机制。方法将15只大鼠分为假手术组(A组)、CIS组(B组)和食欲肽组(C组),各5只。建模前,C组大鼠腹腔注射30 g/kg食欲肽,A组、B组大鼠灌胃等量生理盐水,每日1次,连续7 d。B组和C组大鼠构建短暂性大脑中动脉闭塞(t-MCAO)再灌注模型。采用连续脑电图评估大鼠的睡眠功能,采用转棒实验及爬梯实验评估行为学功能,采用氧化三苯四氮唑(TTC)染色法评估大鼠的脑梗死面积,采用免疫荧光染色法评估大鼠的海马神经元凋亡情况。结果与B组比较,C组大鼠清醒时脑电α波幅度、快速眼动睡眠(REM)期θ波幅、非快速眼动睡眠(NREM)期θ波幅均增高,REM期平均持续时长明显缩短(P<0.01);C组大鼠的脑梗死面积明显缩小(P<0.01);C组大鼠的海马区神经元数量明显增加(P<0.01),凋亡神经元数量明显减少(P<0.01);C组大鼠的爬行错误步数评分明显降低(P<0.01),转棒停留时间明显延长(P<0.01)。结论食欲肽对CIS模型大鼠的神经元有保护作用,且可改善运动功能及运动-感觉功能。Objective To investigate the protective effect and mechanism of orexin on cerebral ischemic stroke(CIS)model rats.Methods Fifteen rats were divided into the sham operation group(group A),CIS group(group B),and orexin group(group C),with five rats in each group.Before establishing the models,rats in group C were intraperitoneally injected with 30 g/kg of orexin,while rats in groups A and B received intragastric administration of an equal volume of physiological saline.The rats in groups B and C were used for establishing transient middle cerebral artery occlusion(t-MCAO)reperfusion models.Continuous electroencephalogram(EEG)was used to evaluate the sleep function of rats,and the rotating rod test and climbing ladder test were used to evaluate behavioral function.TTC staining was used to evaluate the infarct area of rats,and immunofluorescence staining was used to evaluate the apoptosis of hippocampal neurons.Results Compared with those in group B,the amplitude of EEGαwaves,rapid eye movement(REM)phase θ waves,and non-rapid eye movement(NREM)phase θ waves in group C increased during wakefulness,while the average duration of REM phase in group C was significantly shorter(P<0.01).Compared with those in group B,the cerebral infarction area in group C significantly decreased(P<0.01),the number of neurons in the hippocampus in group C significantly increased(P<0.01),the number of apoptotic neurons in group C significantly decreased(P<0.01);the crawling error step count score in group C significantly decreased(P<0.01),while the stick turning dwell time in group C significantly prolonged(P<0.01).Conclusion Orexin has a protective effect on neurons in CIS model rats,which can improve motor function and motor-sensory function.
