摘　　要：目的建立同时测定抗瘤丸(活血益气方)中10种活性成分含量的超高效液相色谱(UPLC)法。方法色谱柱为Waters Acquity BEH C18柱(50 mm×2.1 mm,1.7μm),流动相为水-乙腈(梯度洗脱),流速为0.3 mL/min,检测波长为220 nm(山柰酚、落新妇苷、槲皮素、大黄酸、大黄素、大黄素甲醚)和203 nm(人参皂苷Rg1、人参皂苷Rb1、三七皂苷R1、薯蓣皂苷),柱温为40℃,进样量为2μL。结果10种活性成分在各自质量浓度范围内与峰面积线性关系良好(r≥0.9995,n=7);精密度、稳定性、重复性试验结果的RSD均小于4.0%(n=6);平均加样回收率为89.02%~94.65%,RSD为0.66%~8.70%(n=6)。样品中10种活性成分的平均含量为0.07~7.32 mg/g(n=3)。结论该方法操作简便、重复性好、结果准确,可用于同时测定抗瘤丸(活血益气方)中10种活性成分的含量。Objective To establish an ultra-high performance liquid chromatography(UPLC)method for simultaneous determination of 10 active ingredients in Kangliu Pills(Huoxue Yiqi Formula).Methods The chromatographic column was Waters Acquity BEH C18 column(50 mm×2.1 mm,1.7μm),the mobile phase was water-acetonitrile(gradient elution),the flow rate was 0.3 mL/min,the detection wavelength was set at 220 nm(kaempferol,taxifolin 3-O-rhamnoside,quercetin,rhein,emodin,emodin-3-methll ether)and 203 nm(ginsenoside Rg1,ginsenoside Rb1,notoginsenoside R1,dioscin),the column temperature was 40℃,and the injection volume was 2μL.Results The 10 active ingredients showed good linear relationship with peak areas within their respective mass concentration ranges(r≥0.9995,n=7).The RSDs of precision,stability,and repeatability test results were all lower than 4.0%(n=6).The average recoveries of 10 active ingredients were in the range of 89.02%-94.65%,with RSDs of 0.66%-8.70%(n=6).The average content of 10 active ingredients in the sample was in the range of 0.07-7.32 mg/g(n=3).Conclusion The method is simple,reproducible and accurate,which can be used for simultaneously determination of 10 active ingredients in Kangliu Pills(Huoxue Yiqi Formula).
