摘 要:目的:研究枳实不同生长时期的转录水平差异,探究调控枳实有效成分生物合成的基因。方法:利用Illumina NovaSeq 6000高通量测序技术对不同发育时期的枳实进行转录组测序,利用DESeq筛选差异基因,对差异基因进行聚类分析,将差异基因进行GO富集分析和KEGG富集分析。结果:共获得517950818条clean reads,筛选发现有15064个基因至少在两个时期差异表达,花期后75 d枳壳和花期后30、45、60 d的枳实差异表达基因主要富集到苯丙素合成、类黄酮合成等代谢通路。结论:在类黄酮生物合成通路中,大部分基因都在花期后60 d高表达,从分子层面解释了枳实中总黄酮含量高于枳壳的原因,说明此时枳实入药效果最好。Objective:To study the difference of transcriptional level of Aurantii Fructus Immaturus in different growth stage and explore genes which regulate active constituent biosynthesis.Methods:Illumina NovaSeq 6000 high-throughput sequencing technology was used to conduct transcriptome sequencing for different developmental stages of Aurantii Fructus Immaturus.Differential expressed genes were selected by DESeq and analyzed by cluster analysis,GO enrichment,KEGG enrichment.Results:A total of 517950818 clean reads were acquired and a total of 15064 genes were found to be differentially expressed in at least two periods.The differentially expressed genes of Aurantii Fructus at 75 days after flowering and Aurantii Fructus Immaturus at 30,45 and 60 days after flowering were mainly enriched in phenylpropanoid biosynthesis and flavonoid biosynthesis,etc..Conclusion:In the flavonoid biosynthesis pathway,most genes are highly expressed at 60 days after flowering,which explaines the reason that the total flavonoid content in Aurantii Fructus Immaturus is higher than that in Aurantii Fructus from the molecular level,indicating the best time for drug application.
