摘 要:目的建立微生物快速富集系统(CPS)微生物快速富集系统富集生活污水中的诺如病毒方法,以便应用于生活污水诺如病毒的检测。方法2022年1月1日-2023年12月31日采集梅州市生活污水样品16份,采用CPS微生物快速富集系统进行病毒富集,使用一次性浓缩移液管尖端和湿泡沫洗脱罐,将生物颗粒从大液体体积浓缩到体积为200μL的体积,然后利用磁珠法提取病毒RNA,进行GI、GII型诺如病毒实时荧光定量PCR检测。结果16份生活污水样品富集前后GI、GII型诺如病毒检测结果均为阳性,采用CPS微生物快速富集系统对样品进行前处理后,平均富集效率分别为1.7%和1.8%。两种病毒富集前后的Ct值差异均无统计学意义(P均>0.05)。结论本方法具有一定的富集效率,可应用于生活污水中诺如病毒的检测,但仍需进一步优化前处理。Objective To establish concentrating pipette select(CPS)microbial rapid enrichment system method for enrichment of Norovirus in domestic wastewater,so as to applicate for detecting Norovirus in domestic wastewater.Methods There were 16 domestic wastewater samples collected from Meizhou city from January 1,2022 to December 31,2023.The virus was enriched by CPS microbial rapid enrichment system,and then virus RNA was extracted by magnetic bead method,and the GI and GII type Norovirus in the wastewater samples were detected with real⁃time fluorescence quantification PCR method.Results Norovirus was detected in all 16 domestic sewage samples.And the average enrichment efficiency of GI and GII type Norovirus was 1.7%and 1.8%,respectively,by pretreating with the CPS microbial rapid enrichment system.There was no significant difference in the Ct values before and after the viruses were enriched(both P>0.05).Conclusions The effect of this enrichment method was good,and could be applied to the detection of Norovirus in domestic sewage,but further optimization of pretreatment methods would still be needed to enhance the effect of virus enrichment.
