摘 要:为探究添加不同外源诱导子对牛樟芝菌丝体生长特性的影响,以PDA基础培养基为对照,通过添加不同浓度(50、100、150、200μmol/L)的诱导子茉莉酸甲酯(MeJA)、水杨酸(SA)和油酸(OA)对牛樟芝进行皿式培养,比较分析30 d培养周期中的菌丝体菌落直径、生长速度、生长指数和生物量的差异,筛选出促进牛樟芝生长的适宜诱导子与较优浓度。结果表明:与PDA对照培养基相比,添加3种外源诱导子在各自适宜浓度下对牛樟芝菌丝体的生长均起到显著的促进作用。其中,100μmol/L MeJA条件下的菌落直径达到83.94 mm,最快生长速度为6.17 mm/d,干物质重量为88.10 mg,而150μmol/L MeJA条件下的菌丝体颜色与野生牛樟芝子实体颜色最接近,为棕黄色或棕红色;50μmol/L SA条件下的菌丝体平均生长速度为1.64 mm/d,菌丝体颜色为橘黄色或橘红色,菌落直径为47.5 mm,干物质重量为85.37 mg;添加150和200μmol/L OA条件下的菌丝体颜色为橘黄色或橘红色,菌落直径为43和42.15 mm,平均生长速度为1.43和1.41 mm/d,干物质重量为85.90和79.19 mg。因此,筛选出促进牛樟芝菌丝体生长的适宜外源诱导子为100~150μmol/L MeJA。To investigate the effects of the addition of different exogenous inducers on the growth characteristics of Taiwanofungus camphoratus mycelium,PDA basal medium was used as the control,and T.camphoratus was cultured in plate culture supplemented with different concentrations(50,100,150,or 200μmol·L^(-1))of the inducers methyl jasmonate(MeJA),salicylic acid(SA)and oleic acid(OA).The differences in mycelial colony diameter,growth rate,growth index,and biomass of mycelia were comparatively analyzed during the 30 d culture cycle,and suitable inducers and better concentrations were screened to promote the growth of T.camphoratus.Compared with the PDA control medium,the addition of the three exogenous inducers promoted the growth of the mycelia of T.camphoratus at their respective concentrations.Among them,the colony diameter under 100μmol·L^(-1)MeJA reached 83.94 mm,the fastest growth rate was 6.17 mm·d^(-1),and the dry matter weight was 88.10 mg,while the color of the mycelia under 150μmol·L^(-1)MeJA was the closest to that of the wild T.camphoratus fruiting bodies,which were brownish yellow or brownish red.The average growth rate of mycelia under 50μmol·L^(-1)SA was 1.64 mm·d^(-1),the color of the mycelium was orange or red-orange,the colony diameter was 47.5 mm,and the dry matter weight was 85.37 mg.The color of mycelium under 150μmol·L^(-1)and 200μmol·L^(-1)OA was orange or red-orange,the colony diameters were 43 and 42.15 mm,the average growth rates were 1.43 and 1.41 mm·d^(-1),and the dry matter weights were 85.37 and 79.19 mg,respectively.In this study,a suitable exogenous inducer to promote the growth of the mycelia of the T.camphoratus was determined to be 100-150μmol·L^(-1)MeJA.
