新疆3个地区儿童头癣患者犬小孢子菌的群体遗传学研究

摘  要:目的研究新疆3个(和田、阿克苏及乌鲁木齐)地区儿童头癣患者犬小孢子菌菌株的遗传多样性水平以及菌种的种群结构。方法收集2000年1月-2020年12月新疆3个地区的52株儿童头癣患者的犬小孢子菌菌株,基于微卫星方法,用Popgen软件分析观察等位基因数、有效等位基因数、香农信息指数、Nei氏基因多样性指数。采用GenAlEx软件进行分子方差变异分析和主坐标分析;采用NTSYS软件构建聚类图;采用STUCTURE软件分析犬小孢子菌的种群结构。结果和田地区犬小孢子菌的观察等位基因数为2.091,有效等位基因数为1.878,Nei氏基因多样性指数和Shannon信息指数分别为0.307和0.596,阿克苏地区犬小孢子菌的观察等位基因数为2.182,有效等位基因数为1.503,Nei氏基因多样性指数和Shannon信息指数为0.307和0.457,乌鲁木齐地区犬小孢子菌的观察等位基因数最高达到3.000,有效等位基因数为1.447,Nei氏基因多样性指数和Shannon信息指数分别为0.307和0.492。种群间遗传变异占4%,种群内遗传变异占96%。阿克苏地区犬小孢子菌和乌鲁木齐地区犬小孢子菌遗传距离最小,遗传相似度最大,亲缘关系较近,和田地区犬小孢子菌与阿克苏地区犬小孢子菌遗传距离最大,遗传相似度最小,新疆3个地区犬小孢子菌可被分为3个谱系。结论新疆和田、阿克苏及乌鲁木齐3个地区的儿童头癣患者的犬小孢子菌具有遗传多样性。Objective Study the genetic diversity level and population structure of Microsporum canis strains in children with tinea capitis in three regions of Xinjiang(Hotan,Aksu and Urumqi).Methods 52 strains of Microsporum canis(M.canis)from children with tinea capitis were collected from January 2000 to December 2020 in the 3 regions of Xinjiang,and theywere analysed by Popgen software to observe the number of alleles,the number of effective alleles,Shannon's information index and Nei's unbiased gene diversity index,GenAlEx software was used to perform molecular variance analysis of variance and principal coordinates analysis.NTSYS software was used to construct a NTSYS software was used to construct the cluster diagram.STUCTURE software was used to analyse the population structure of Microsporum ca-nis.Results The observed allele number of M.canis in Hotan was 2.091,the effective allele number was 1.878,and the Nei's gene diversity index and Shann on information index were 0.307 and 0.596,respec-tively;the observed allele number of M.canis in Aksu was 2.182,the effective allele number was 1.503,and the Nei gene diversity index and Shannon information index were also 0.307 and 0.596,respectively.Shannon's information index was also 0.307 and 0.457.The highest observed allele number of M.canis in Urumqi was 3.000,the effective allele number was 1.447,and the Nei's gene diversity index and Shan-non's information index were 0.307 and 0.492,respectively.Inter-population genetic variation accounted for 4%,and intra-population genetic variation accounted for 96%.The genetic distance between M.canis in Aksu region and M.canis in Urumqi region was the smallest,the genetic similarity was the largest,and the relatives were closer,and the genetic distance between M.canis in Hotan region and M.canis in Aksu region was the largest,and the genetic similarity was the smallest,and the M.canis in the 3 regions in Xinjiang can be classified into 3 lineages.Conclusion There is genetic diversity of M.canis in children with tinea capitis in
