摘 要:目的开发一款可检测所有Aβ聚集体,并具有序列选择性的长寿命Aβ荧光探针(Ru-itatp)。方法利用荧光光谱测试Ru-itatp与Aβ蛋白的作用及荧光检测效果,利用MTT法测试生物毒性,利用激光共聚焦显微成像研究细胞及组织成像效果。结果Ru-itatp对Aβ_(1-42)蛋白有灵敏的荧光强度和荧光寿命响应,可与不同聚集形态(单体、低聚体、纤维体)Aβ结合,能特异性标记大脑中的Aβ斑块,并具有较低的生物毒性。结论Ru-itatp具有较强的Aβ蛋白识别能力及较低的生物毒性,有望成为阿尔茨海默症早期诊断和治疗的有力工具。Objective To develop a highly selective Aβfluorescent probe that can detect all Aβaggregated species with sequence recognition.Methods The interaction of Ru-itatp with Aβprotein and fluorescence detection were tested by fluorescence spectroscopy,its biotoxicity was tested by MTT method,and its cellular and tissue imaging effect was investigated by laser confocal microimaging.Results Ru-itatp had a sensitive fluorescence intensity and fluorescence lifetime response to Aβ_(1-42)proteins,binded to Aβin different aggregated forms(monomers,oligomers,and fibrils),and specifically labeled Aβplaques in the brain with low biotoxicity.Conclusion Ru-itatp,with its strong Aβprotein recognition ability and low biotoxicity,is expected to be a powerful tool for early diagnosis and treatment of Alzheimer’s disease.
