经典名方保元汤UPLC特征图谱研究

摘　　要：目的 建立经典名方保元汤的UPLC特征图谱,为其质量标准的建立提供参考。方法 色谱柱为Agilent ZORBAX SB C18(2.1 mm×150 mm,1.8μm);流动相为乙腈-水(梯度洗脱),流速为0.20 mL/min,柱温为35℃,进样量为1μL,检测波长为203 nm。以峰5芒柄花苷为参照峰,绘制21批保元汤基准样品的UPLC特征图谱,采用《中药色谱指纹图谱相似度评价系统(2012版)》进行相似度评价。结果 21批保元汤基准样品共有12个特征峰,分别为毛蕊异黄酮葡萄糖苷(峰1)、新甘草苷(峰2)、甘草苷(峰3)、芹糖异甘草苷(峰4)、芒柄花苷(峰5)、异甘草苷(峰6)、甘草素(峰7)、人参皂苷Re(峰8)、人参皂苷Rg1(峰9)、毛蕊异黄酮(峰10)、芒柄花素(峰11)、人参皂苷Rd(峰12),相似度大于0.70。结论 所建立的UPLC特征图谱和特征峰的分析结果可为经典名方保元汤的质量控制提供参考。Objective To establish the UPLC characteristic chromatogram of the famous classical formula Baoyuan Decoction and provide reference for the establishment of its quality standard.Methods The determination was performed on Agilent ZORBAX SB C18(2.1 mm×150 mm,1.8μm)column with mobile phase consisted of acetonitrile-water(gradient elution)at the flow rate of 0.20 mL/min.The column temperature was 35℃and the sample size was 1μL.The detection wavelength was set at 203 nm.The UPLC characteristic chromatograms of 21 batches of substance benchmark samples of Baoyuan Decoction were plotted with peak 5,ononin,as the reference peak,and the similarity evaluation was carried out by the Similarity Evaluation System of Chromatographic Fingerprint of Traditional Chinese Medicine(2012 Edition).Results There were 12 characteristic peaks in the 21 batches of the substance benchmark samples of the Baoyuan decoction,which were galycosin-7-O-beta-Dglucoside,neoiliquiritin,iliquiritin,isoliquiritin apioside,ononin,isoliquiritoside,liquiritigenin,ginsenoside Re,ginsenoside Rg1,calycosin,formononetin,ginsenoside Rd,with the degree of similarity of more than 0.70.Conclusion The results of the established UPLC characteristic chromatogram and characteristic peak correlation analysis can provide a reference for the quality control of Baoyuan Decoction.
