摘  要:目的 建立同时测定黄芪中多指标成分含量的高效液相色谱方法,测定宁夏不同生长期栽培黄芪的多指标成分,并结合化学计量学分析,对宁夏栽培黄芪的质量进行评价。方法 采用高效液相色谱-二极管阵列检测器-蒸发光检测器(HPLC-DAD-ELSD)同时测定毛蕊异黄酮葡萄糖苷、芒柄花苷、芒柄花素、毛蕊异黄酮、黄芪甲苷、黄芪皂苷Ⅲ、黄芪皂苷Ⅱ、黄芪皂苷Ⅰ、异黄芪皂苷Ⅱ和异黄芪皂苷Ⅰ的含量;利用Origin Pro2022软件采用偏最小二乘判别分析、主成分分析和聚类分析手段,分析宁夏栽培黄芪中各指标成分随生长年限的变化规律。结果 测定了34批不同生长期宁夏栽培黄芪中10个成分的含量,偏最小二乘-判别分析表明毛蕊异黄酮葡萄糖苷、毛蕊异黄酮、芒柄花素、黄芪甲苷、黄芪皂苷Ⅰ为代表不同生长期黄芪的主要特征成分。聚类分析显示按照生长年限不同,宁夏栽培黄芪基本可聚为5类。结论 建立了HPLC-DAD-ELSD含量测定方法,该方法测定结果表明宁夏不同生长期栽培黄芪中成分的积累受种植年限、种植地域的双重影响,生长年限的增加和砂质土壤均有益于黄芪成分的积累。Objective To establish a high performance liquid chromatography(HPLC)method for the simultaneous determination of the contents of multiple index components in Astragalus,measure the multiple indicator components of cultivated Astragalus in Ningxia at different growth periods,and combine with chemometrics analysis to evaluate the quality of cultivated Astragalus in Ningxia.Methods HPLC-DAD-ELSD method was used to simultaneously determine the contents of mauve isoflavone glucoside,ononin,mangiferin,mauve isoflavone,astragaloside,astragalosideⅢ,astragalosideⅡ,astragalosideⅠ,isoastragalosideⅠ,isoastragalosideⅡ.The Origin Pro 2022 software was used,the partial least squares discriminant analysis,principal component analysis,and cluster analysis methods were employed to analyze the variation law of each indicator component in the cultivated Astragalus with the growth period.Results The contents of 10 components in 34 batches of Astragalus at different growth stages in Ningxia were determined,and the partial least squares discriminant analysis showed that five components,namely,trichostatin glucoside,trichostatin isoflavone,mangostin,astragaloside,astragalosideⅠ,were the main components representing the different growth stages of Astragalus.Cluster analysis showed that according to the different growth periods,the cultivated Astragalus in Ningxia could be basically divided into five categories.Conclusion This study established an HPLC-DAD-ELSD content determination method.The measurement results showed that the accumulation of components in the cultivated Astragalus in Ningxia at different growth periods was affected by the dual influence of planting years and planting regions,and the increase in growth years and sandy soil were both beneficial to the accumulation of Astragalus components.
