摘 要:为探究野生酸模中草酸提取的条件及测定方法的建立,以野生酸模为试验材料,以盐酸为提取剂,研究磺基水杨酸铁络合物比色法和甲基红-重铬酸钾催化光度法测定草酸含量的差异。结果表明,影响草酸提取因素主次顺序为料液比>浸提时间>盐酸浓度,且料液比、浸提时间影响显著,盐酸浓度影响不显著。草酸提取的最佳条件为料液比2∶15,提取时间15 min,盐酸浓度0.60 mol/mL,在此条件下,草酸提取量达1.71 g/100 g。磺基水杨酸铁络合物比色法加标回收率为88.4%~103.9%,最小检出限为0.14μg/mL,精密度为90.7%~102.5%;甲基红-重铬酸钾催化光度法加标回收率为81.7%~96.7%,最小检出限为0.03μg/mL,精密度为83.0%~94.3%。磺基水杨酸铁络合物比色法测定的精密度、最小检出限均高于甲基红-重铬酸钾催化光度法,前者比后者更加适宜于野生酸模中草酸含量的测定。In order to explore the conditions for extracting oxalic acid from wild Rumex and establish a determination method,wild Rumex was used as experimental materials and hydrochloric acid was used as the extraction agent to study the differences in determining oxalic acid content between sulfosalicylic acid iron complex colorimetric method and methyl red potassium dichromate catalytic photometric method.The results showed that the primary and secondary factors affecting oxalic acid extraction were in the order of solid-liquid ratio>extraction time>hydrochloric acid concentration,and the influence of solid-liquid ratio and extraction time was significant,while the influence of hydrochloric acid concentration was not significant.The optimal conditions for oxalic acid extraction were solid-liquid ratio of 2∶15,extraction time of 15 min,and hydrochloric acid concentration of 0.60 mol/mL.Under these conditions,the oxalic acid extraction yield reaches 1.71 g/100 g.The colorimetric recovery rate of sulfosalicylic acid iron complex was between 88.4%and 103.9%,with a minimum detection limit of 0.14μg/mL and a precision was 90.7%~102.5%;The recovery rate of methyl red potassium dichromate catalytic spectrophotometric method was 81.7%~96.7%,with a minimum detection limit of 0.03μg/mL and a precision of 83.0%~94.3%.The precision and minimum detection limit of the sulfosalicylic acid iron complex colorimetric method were higher than those of the methyl red potassium dichromate catalytic photometric method,and the former was more suitable for the determination of oxalic acid content in wild Rumex than the latter.
