基于网络药理学探讨痰热清注射液治疗急性呼吸窘迫综合征的作用机制

摘　　要：目的基于网络药理学探讨痰热清注射液治疗急性呼吸窘迫综合征(ARDS)的作用机制。方法通过TCMSP、BATMAN数据库检索痰热清注射液的活性成分及作用靶点,利用OMIM、Genecards数据库筛选出ARDS的疾病潜在靶点,将药物及疾病靶点进行映射,运用Cytoscape3.9.0构建“药物—成分—靶点”网络图,筛选关键靶点。运用STRING数据库构建蛋白相互作用,对潜在靶点进行GO及KEGG富集分析。结果通过筛选共获得痰热清注射液有效活性成分934个,疾病靶点374个,与ARDS共同靶点67个。GO富集分析得到1643个生物学过程,KEGG通路富集得到176条信号通路,与ARDS相关的通路主要有高级糖基化终末产物—受体(AGE-RAGE)信号通路、缺氧诱导因子-1(HIF-1)信号通路、肿瘤坏死因子(TNF)信号通路、白介素-17(IL-17)信号通路。结论通过网络药理学技术分析,痰热清注射液可能通过参与血管内皮保护作用相关通路发挥治疗ARDS的作用。Objective To investigate the mechanism of Tanreqing injection in the treatment of acute respiratory distress syndrome(ARDS)based on network pharmacology.Methods The active ingredients and target of Tanreqing injection were searched using TCMSP and BATMAN databases,potential disease targets of ARDS were screened using OMIM and Genecards databases.Drug and disease targets were mapped,and the"drug-components-target"network diagram was constructed using Cytoscape3.9.0.screening key targets.Protein interactions were constructed using STRING database,GO and KEGG enrichment analysis were performed for potential targets.Results A total of 934 active ingredients of Tanreqing injection,374 disease targets and 67 common targets with ARDS were obtained.GO enrichment analysis yielded 1643 biological processes and KEGG enrichment yielded 176 signaling pathways.Advanced glycation end-product receptor(AGE-RAGE)signaling pathway,hypoxia-inducing factor-1(HIF-1)signaling pathway,tumor necrosis factor(TNF)signaling pathway,and interleukin-17(IL-17)signaling pathway were mainly involved in ARDS.Conclusion Through the analysis of network pharmacology,Tanreqing injection may play a role in the treatment of ARDS by participating in the vascular endothelial protective pathways.
