摘 要:目的对桑叶中1-脱氧野尻霉素(1-deoxynojirimycin,DNJ)进行提取与定量后研究其对结肠癌HT-29细胞及其移植瘤小鼠的抗肿瘤作用。方法测定DNJ对HT-29细胞增殖和凋亡情况的影响。用HT-29细胞悬液制备裸鼠结肠癌皮下移植瘤模型,并将造模成功的模型鼠随机分为模型对照组、低剂量DNJ组和高剂量DNJ组,给药结束后测定各组小鼠肿瘤体积、肿瘤质量和抑瘤率,最后用TUNEL染色法测定小鼠肿瘤组织的细胞凋亡情况。结果DNJ在高效液相色谱中的出峰时间为12.52 min,对HT-29细胞增殖具有抑制作用和促其凋亡的作用。在裸鼠皮下移植瘤模型中,高浓度DNJ显著减小肿瘤体积和质量,抑瘤率达到32.85%。DNJ还能诱导小鼠肿瘤细胞发生凋亡,且高浓度DNJ作用更加明显。结论DNJ对HT-29细胞及其小鼠移植瘤具有抗肿瘤作用。OBJECTIVE To extract and quantify 1-deoxynojirimycin(DNJ)from mulberry leaves and study its anti-tumor effect on colon cancer HT-29 cells and transplanted tumor mice.METHODS The effects of DNJ on the proliferation and apoptosis of HT-29 cells were determined.Subcutaneous transplantation model of colon cancer in nude mice was established by HT-29 cell suspension.The successful model mice were randomly divided into three groups:model control group,low dose DNJ group and high dose DNJ group.The tumor volume,tumor weight and tumor inhibition rate of each group were measured after the end of treatment.Finally,the apoptosis of tumor tissue was detected by TUNEL staining.RESULTS The peak time of DNJ in high performance liquid chromatography was 12.52 min.DNJ could inhibit the proliferation of HT-29 cells and promote their apoptosis.In the subcutaneous transplanted tumor model of nude mice,high concentration of DNJ significantly reduced tumor volume and mass,and the tumor inhibition rate reached 32.85%.DNJ also induced apoptosis of tumor cells in mice,and the effect was more obvious at high concentration of DNJ.CONCLUSION DNJ showed anti-tumor effect on HT-29 cells and its transplanted tumor mice.
