17-β雌二醇对大鼠机械性通气肺损伤的肺保护作用的研究

摘　　要：目的研究17-β雌二醇对大鼠机械性通气肺损伤(VILI)的肺保护作用。方法27只健康雄性SD大鼠随机分为3组:T1(小潮气量组),VT=6 mL·kg^(-1);T2(大潮气量组),VT=20 m1·kg^(-1);T3(大潮气量+17-β雌二醇组),VT=20 mL·kg^(-1)。机械通气前2 h给予17-β雌二醇(E2:50 g·kg^(-1))灌胃。各组通气时间2 h。结果灌胃17-β雌二醇虽然没有明显降低大鼠肺组织的含水量。但是,HE染色结果表明T3(大潮气量+17-β雌二醇组)相较于T2(大潮气量组),病理状况有改善。ELISA结果表明大鼠灌胃17-β雌二醇可显著降低促炎因子IL-1β的含量,可明显提高抑炎因子IL-10的含量(P≤0.05)。结论大鼠灌胃17-β雌二醇可降低大潮气量机械通气对大鼠肺组织造成的炎症和氧化损伤,可以推测该药物对大鼠机械性通气肺损伤具有一定的保护作用。OBJECTIVE To study the protective effect of 17-βestradiol on mechanical ventilation lung injury(VILI)in rats.METHODS Twenty-seven healthy male SD rats were randomly divided into 3 groups:T1(small tidal volume group),VT=6 mL·kg^(-1);T2(high tidal volume group),VT=20 m1·kg^(-1);T3(high tidal volume+17-βestradiol group),VT=20 mL·kg^(-1).17-βestradiol(E2:50 g·kg^(-1))was intragastric administration 2 h before mechanical ventilation.The ventilation time of each group was 2 h.RESULTS Intragastric administration of 17-βestradiol could not significantly reduce the water content of lung tissue.However,HE staining results showed that T3(high tidal volume+17-βestradiol group)had improved pathological status compared with T2(high tidal volume group).ELISA results showed that intragastric administration of 17-βestradiol could significantly reduce the content of pro-inflammatory factor IL-1β,and significantly increase the content of anti-inflammatory factor IL-10(P≤0.05).CONCLUSION Intragastric administration of 17-βestradiol can reduce the inflammation and oxidative damage of lung tissue induced by mechanical ventilation of high tidal volume.It can be inferred that 17-βestradiol has a certain protective effect on mechanical ventilation lung injury in rats.
