摘 要:为了探究细胞外信号调节激酶2(extracellular signal-regulated kinase 2,ERK2)在性成熟前绵羊不同生殖器官中的表达差异,揭示绵羊生殖器官发育调控机制,试验选取出生日期一致、健康的性成熟前(3月龄)的小尾寒羊雌、雄性羔羊各3只,取子宫、卵巢、输卵管及睾丸、附睾、输精管组织样品,采用荧光定量PCR方法检测ERK2基因的相对表达量,采用免疫组织化学方法检测ERK2蛋白在绵羊生殖器官的表达定位情况,采用Western-blot方法检测ERK2蛋白在绵羊各生殖器官的表达量。结果表明:雌性绵羊各生殖器官ERK2基因相对表达量均差异不显著(P>0.05);雄性绵羊生殖器官中,睾丸ERK2基因相对表达量极显著高于附睾和输精管(P<0.01),附睾和输精管之间差异不显著(P>0.05)。ERK2蛋白在性成熟前绵羊各生殖器官中均有表达。雌性绵羊子宫ERK2蛋白表达量极显著高于输卵管和卵巢(P<0.01),卵巢和输卵管之间差异不显著(P>0.05);雄性绵羊睾丸ERK2蛋白表达量极显著高于附睾和输精管(P<0.01),附睾显著高于输精管(P<0.05)。说明ERK2在性成熟前绵羊各生殖器官中均有表达,但表达量具有差异,其中雌性以子宫表达量居多,雄性以睾丸表达量居多。In order to explore the differential expression of extracellular signal-regulated kinase 2(ERK2)in different reproductive organs of prepubertal sheep and shed light on the regulatory mechanisms of reproductive organ development in sheep,a study was conducted using 3 healthy prepubertal(3 months of age)small-tailed Han sheep lambs of each sex with the same birth date.Tissue samples were collected from uterus,ovary,and fallopian tube as well as the testis,epididymis and vas ductus.The relative expression levels of ERK2 gene was detected by fluorescence quantitative PCR method.The expression and location of ERK2 protein in reproductive organs of sheep were detected by immunohistochemical method,and the expression levels of ERK2 protein in reproductive organs of sheep was detected by Western-blot method.The results showed that there were no significant differences in the relative expression levels of ERK2 gene in the reproductive organs of female sheep(P>0.05).In the reproductive organs of male sheep,the relative expression level of ERK2 gene in testis was significantly higher than that in the epididymis and vas deferens(P<0.01),but there was no significant difference between the epididymis and vas deferens(P>0.05).ERK2 protein was expressed in all reproductive organs of prepubertal sheep.The expression level of ERK2 protein in uterus of female sheep was significantly higher than that in the fallopian tube and ovary(P<0.01),with no significant difference between the ovary and fallopian tube(P>0.05).The expression level of ERK2 protein in male sheep testis was significantly higher than that in epididymis and vas deferens(P<0.01),and epididymis was significantly higher than in that the vas deferens(P<0.05).These results indicated that ERK2 was expressed in all reproductive organs of sheep before sexual maturity,but the expression levels varied,with uterus showing the highest expression in females and the testes showing the highest expression in males.
