摘 要:目的建立波长转换法测定威灵仙中的异阿魏酸、灵仙新苷含量。方法采用反相高相液相色谱法,色谱柱为CAPCELLPAK MG C_(18)(4.6 mm×250 mm,5μm)。流动相A为乙腈,B为0.1%磷酸溶液,梯度洗脱,流速为1.0 ml/min,检测波长:316 nm(异阿魏酸),205 nm(灵仙新苷)。结果异阿魏酸、灵仙新苷的浓度分别在0.8215~12.3231、170.91~1367.30μg/ml的范围内呈线性关系(r=0.9999、0.9997);异阿魏酸、灵仙新苷平均加样回收率分别为97.41%(n=6)、96.40%(n=6)。结论所建立的测定方法操作简便、灵敏度高。Objective To establish a wavelength conversion method to determine the content of isoferulic acid and Lingxianxin glycoside in Weilingxian.Methods Reverse phase high-performance liquid chromatography was used,with chromatographic column of CAPCELLPAK MG C18(4.6 mm×2500 mm,5μm).The mobile phase A was acetonitrile,and B was 0.1%phosphoric acid solution.The gradient elution was performed with the flow rate of 1.0 ml/min,and the detection wavelength was 316 nm(isoferulic acid)and 205 nm(Lingxianxin glycoside).Results The concentrations of isoferulic acid and Lingxianxin glycoside showed a linear relationship within the ranges of 0.8215 to 12.3231 and 170.91 to 1367.30μg/ml,respectively(r=0.9999,0.9997);The average recovery rates of isoferulic acid and Lingxianxin glycoside were 97.41%(n=6)and 96.40%(n=6),respectively.Conclusion The established measurement method is easy to operate and has high sensitivity.
