摘 要:板角山羊(Banjiao goat)和酉州乌羊(Youzhou dark goat)是重庆地方特色品种山羊(Capra hircus)。为研究板角山羊和酉州乌羊基因的差异表达,本研究利用Illumina测序技术分别对板角山羊和酉州乌羊背最长肌、心脏和肝脏进行转录组测序,对测序结果分别进行序列比对、结构分析和转录因子预测,筛选差异表达基因(differentially expressed genes,DEGs)并对其进行功能富集分析,并随机选取6个DEGs进行RT-qPCR验证。结果表明,在板角山羊和酉州乌羊的心脏、背最长肌和肝脏组织中检测出1390854个单核苷酸多态性(single nucleotide polymorphism,SNP)位点、74758个插入/缺失(insertion/deletion,Indel)和53625个可变剪接,鉴定了66、190和377个DEGs,这些基因被划分到14种转录因子家族中,同时参与了与肌肉组成成分、有机酸及脂肪酸代谢等相关的多个代谢通路。本研究发现,ASIP基因在山羊体内可能通过参与调节不同组织的脂肪代谢进而影响肉的品质,ANKRD1基因可能是另一个影响板角山羊和酉州乌羊肉品质差异的重要候选基因,DNAJB1、MTL5和N2DL2基因可能参与了不同品种山羊的免疫机能调节。本研究阐述了板角山羊和酉州乌羊不同组织的基因表达差异,为板角山羊和酉州乌羊遗传资源的挖掘与利用提供了重要信息。Banjiao goat and Youzhou dark goat are local characteristic breeds of goats(Capra hircus)in Chongqing.The purpose of this study was to study the gene differential expression of Banjiao goat and Youzhou dark goat.In this study,Ilumina sequencing technology was used to perform transcriptome sequencing on longissimus dorsi muscle,heart,and liver from Banjiao goat and Youzhou dark goat.Sequence alignment,structural analysis,and transcription factor prediction were performed,and differentially expressed genes(DEGs)were screened for functional enrichment analysis.Six DEGs were randomly selected for RT-qPCR verification.Analysis of the results showed that 1390854 single nucleotide polymorphism(SNP)sites,74758 insertion/deletion(Indel),and 53625 alternative splicing were detected in the heart,longissimus dorsi muscle,and liver tissues from Banjiao goat and Youzhou dark goat.66,190,and 377 DEGs were identified in dfferential expression analysis,and these genes were divided into 14 transcription factor families.The DEGs were also involved in several metabolic pathways related to muscle composition,organic acid and fatty acid metabolism.This study found that the ASIP gene in goats may affect meat quality by partici-pating in the regulation of fat metabolism in different tissues.The ANKRDI gene could be another important candidate gene that inf-luencing the differences in meat quality between Banjiao goat and Youzhou dark goat.The DNAJBI,MTL5,and N2DL2 gene may be involved in the immune regulation of different breeds of goats.This study elucidates the gene expression difference between Banjiao goat and Youzhou dark goat,which provides important information for the mining and utilization of genetic resources of Banjiao goat and Youzhou dark goat.
