摘 要:为研究黄河上游不同群体大鼻吻鮈遗传结构与系统发育关系,保护其野生种质资源,本研究基于线粒体Cytb和COⅠ基因对黄河宁夏段及内蒙古段4个地理群体的大鼻吻鮈进行了遗传多样性及系统发育关系分析。结果表明,Cytb和COⅠ基因序列长度分别为1145、749 bp,两个序列串联后的组合序列长度为1894 bp,碱基组成均呈现明显的AT偏倚性。4个地理种群中,线粒体Cytb和COⅠ序列具有8个组合多态位点及9个组合单倍型,平均单倍型多样性(H_(d))0.776,平均核苷酸多样性(π)为0.00078。分子方差分析(analysis of molecular variance,AMOVA)显示4个地理种群间无显著遗传分化,其中95.81%变异来自群体内部,4.18%变异来自群体间。系统发育树及单倍型网络图表明大鼻吻鮈遗传结构比较单一,错配分布结果表明大鼻吻鮈在历史上出现过群体扩张事件。综上所述,黄河中上游的大鼻吻鮈群体遗传结构单一、遗传多样性较低,有必要将其作为一个保护单元进行就地保护,以期丰富大鼻吻鮈种群的遗传多样性。In order to study the genetic structure and phylogenetic relationship of Rhinogobio nasutus in different populations in the upper reaches of the Yellow River and protect its wild germplasm resources,the genetic diversity and phylogenetic relationship of four populations in the Ningxia section and Inner Mongolia section of the Yellow River were analyzed based on mitochondrial Cytb and CO I genes.The results showed that the sequence length of Cytb and CO I genes was 1145 bp and 749 bp,respectively,and the combined sequence length of the two sequences was 1894 bp,showing an obvious AT bias in base composition.The mitochondrial Cytb and CO I sequences in the four populations had 8 combined polymorphic sites and 9 combined haplotypes,including a mean haplotype diversity(H_(d))of 0.776 and mean nucleotide diversity(π)of 0.00078.Analysis of molecular variance(AMOVA)showed that there was no significant genetic differentiation among the four populations,and 95.81%of the variation came from within the populations.4.18%of the variation was among the populations.The phylogenetic tree and haplotype network map indicated that the genetic structure of Rhinogobio nasutus was relatively simple,and the results of the mismatch distribution indicated that there had been a population expansion event in the history of Rhinogobio nasutus.In summary,the genetic structure of the population of Rhinogobio nasutus in the middle and upper reaches of the Yellow River is single,and the genetic diversity is low.It is necessary to conserve the population as a whole in situ,so as to enrich the genetic diversity of the population.
