摘  要:目的研究芪楼丸含药血清对人肝癌HepG2细胞的体外抗癌作用及其机制。方法将人肝癌HepG2细胞体外培养,分为对照组,芪楼丸低、中、高剂量组,各组用对应含药血清进行干预。CCK-8法和Annexin V-FITC/PI双染法分别检测HepG2细胞的增殖和凋亡情况。实时荧光定量PCR法和Western blot法分别检测各组细胞中P53、P21、Caspase-3、Bax、Bcl-2 mRNA及蛋白的表达水平。结果(1)芪楼丸含药血清呈浓度及时间依赖性抑制人肝癌HepG2细胞的增殖,与对照组增殖抑制率相比,差异有统计学意义(P<0.01)。(2)芪楼丸含药血清可促进人肝癌HepG2细胞的凋亡,且随药物浓度增加这种促进作用越显著,其中高、中剂量组与对照组相比差异具有统计学意义(P<0.01),而低剂量组差异则不显著(P>0.05)。(3)与对照组相比,芪楼丸含药血清各组P53、P21、Caspase-3、Bax mRNA的表达随药物剂量的增加呈上升趋势,而Bcl-2 mRNA则与之相反,差异具有统计学意义(P<0.05或P<0.01)。(4)相比于对照组,芪楼丸含药血清各剂量组中P53、P21、Caspase-3、Bax蛋白表达水平均有所增加,且随着药物剂量的增加呈上升趋势(P<0.05或P<0.01);相反,Bcl-2蛋白表达水平随着药物剂量的增加逐渐下降,差异有显著性(P<0.01)。结论芪楼丸可通过参与P53信号通路的传导,进而抑制人肝癌HepG2细胞的增殖,促进其凋亡,从而发挥治疗肝癌的作用。Objective To study the anticancer effects of Qilou Pill-containing serum on human hepatocellular carcinoma Hep G2 cells in vitro and its mechanism.Methods Human hepatoma Hep G2 cells were cultured in vitro and divided into the control group and the low,medium and high dose groups of Qilou Pills,and each group was treated with corresponding serum containing drugs.The proliferation and apoptosis of Hep G2 cells were detected by CCK-8 method and Annexin V-FITC/PI double staining.The mRNA and protein expression levels of P53,P21,Caspase-3,Bax and Bcl-2 were detected by real-time fluorescence quantitative PCR and Western blot,respectively.Results(1)Qilou Pill-containing serum inhibited the proliferation of human hepatoma Hep G2 cells in a concentration and time dependent manner,with statistical significance compared with the control group(P<0.01).(2)Qilou Pill drug-containing serum could promote the apoptosis of human hepatoma Hep G2 cells,and the promoting effect was more significant with the increase of drug concentration.The difference between medium and high dose groups and control group was statistically significant(P<0.01),while the difference between low dose group was not significant(P>0.05).(3)Compared to the control group,the mRNA expressions of P53,P21,Caspase-3 and Bax in Qilou Pill serum increased with the increase of drug dose,while the Bcl-2 mRNA showed the opposite trend,with statistical significance(P<0.05 or P<0.01).(4)Compared with the control group,the protein expression levels of P53,P21,Caspase-3 and Bax in Qilou Pill-containing serum dose groups were increased,and showed an increasing trend with the increase of drug dose(P<0.05 or P<0.01).On the contrary,the expression level of Bcl-2 protein decreased gradually with the increase of drug dose,and the difference was statistically significant(P<0.01).Conclusion Qilou Pills can inhibit the proliferation of human hepatoma Hep G2 cells and promote their apoptosis by participating in the transmission of P53 signalling pathway,so as to play a role in th
