摘　　要：目的 采用南方红豆杉果实(TCF)提取物对小鼠进行急性毒性试验,评价TCF的安全性。方法选取雌性、雄性C57BL/6小鼠各20只,根据性别随机分为正常组和TCF组。TCF组按生药量6 g/(kg·d)给予TCF提取物溶液灌胃,正常组按照10 mL/(kg·d)给予生理盐水灌胃,连续给药14 d。观察和记录小鼠行为、精神状态以及死亡情况,每周称重1次。解剖并观察各脏器情况并进行称重,并计算肝脏、脾脏、肾脏、脑指数;检测血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、总蛋白(TP)、白蛋白(ALB)、尿素氮(BUN)、肌酐(CREA)、葡萄糖(GLU)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)、甘油三酯(TG)、总胆固醇(TC)含量;HE染色观察肝、心、肾、大肠、小肠组织病理形态变化。结果 灌胃期间各组小鼠生长发育良好,未出现明显异常行为和中毒症状,未出现死亡。2组不同性别小鼠干预前和干预7、14 d体质量比较,差异均无统计学意义(P>0.05);2组不同性别小鼠肝脏、脾脏、肾脏、脑指数比较,差异均无统计学意义(P>0.05)。2组不同性别小鼠血清ALT、AST、TP、ALB、BUN、CREA、GLU、TG、TC、HDL、LDL含量比较,差异均无统计学意义(P>0.05)。HE染色结果显示:2组小鼠肝脏、肾脏、心脏、大肠、小肠组织形态完整,结构清晰,未发现明显病理变化。结论 TCF在6 g/(kg·d)剂量下,未观察到明显的毒副作用,表明其具有良好的安全性。Objective:To evaluate the safety of Taxus chinensis fruit(TCF)extract by acute toxicity test in mice.Methods:A total of twenty female and twenty male C57BL/6 mice were randomly divided into normal group and TCF group based on gender.The TCF group was given TCF extract solution by gavage at a dose of 6 g/(kg·d),while the normal group was given physiological saline solution by gavage at a dose of 10 mL/(kg·d)for 14 consecutive days.The growth,development and mortality of mice were observed and recorded,and weighed once a week.The organs were dissected and weighed,and the indexes of liver,spleen,kidney and brain were calculated.Serum alanine transaminase(ALT),aspartate transaminase(AST),total protein(TP),albumin(ALB),blood urea nitrogen(BUN),creatinine(CREA),glucose(GLU),high density lipoprotein(HDL),low density lipoprotein(LDL),triglyceride(TG)and total cholesterol(TC)were detected;HE staining was used to observe the pathological changes in liver,heart,kidney,large intestine,and small intestine.Results:During the period of gavage,the mice in each group grew and developed well without obvious abnormal behavior,poisoning symptoms,and death.There was no significant difference in body weight of different genders between the two groups before intervention and on the 7th and 14th day of intervention(P>0.05).There were no significant differences in the indexes of liver,spleen,kidney and brain between the two groups of different genders(P>0.05).HE staining showed that the liver,kidney,heart,large intestine,and small intestine tissues of the two groups were intact and structurally clear,and no obvious pathological changes were found.Conclusion:At a dose of 6 g/(kg·d),no significant toxic side effects were observed with TCF,indicating its good safety.
