摘 要:为探究棕榈拟盘多毛孢(Pestalotiopsis trachicarpicola)病原菌对六妹羊肚菌(Morchella sextelata)生长的影响:检测病原菌对六妹羊肚菌菌丝体的损伤、子囊果的侵染、发酵和挥发性产物对菌丝体生长的影响;研究病原菌侵染过程中羊肚菌的防御酶活性变化、对羊肚菌浸提液的利用情况及细胞壁降解酶活性。结果表明:棕榈拟盘多毛孢攀附缠绕羊肚菌菌丝,导致其皱缩枯萎;与正常子实体相比,病原菌侵染的羊肚菌子囊果组织结构被破坏;病原菌发酵液对羊肚菌菌丝体生长抑制率为7.6%;病原菌与羊肚菌共同培养至第6天时羊肚菌菌丝稀疏,气生菌丝较少;病原菌侵染的子实体SOD活性在第2天时最高,POD活性在第1天时最高,CAT活性在第2天时最低;在实验范围(72 h)内羊肚菌浸提液可促进病原菌分生孢子的萌发;病原菌能产生细胞壁降解酶,在第15天时,中性蛋白酶活性最高,第6天时,β-1,3葡聚糖酶活性最高,在第12天时,几丁质酶活性最高。研究结果可为探究棕榈拟盘多毛孢的致病机制提供参考。The effects of pathogenic Pestalotiopsis trachicarpicola WLM5 on Morchellas extelata were studied in terms of mycelial damage,ascocarp infection,response of mycelia to fermentation and volatile products of P.trachicarpicola,and activities of defense enzymes.Utilization of M.extelata extract by P.trachicarpicola and activities of its cell wall degrading enzymes during infection were also measured.The results showed that hyphae of WLM5 intertwined with those of M.extelata,making them shrivel and wilt.WLM5 infection also resulted in destruction of ascocarp tissue structure.The fermentation liquid of P.trachicarpicola inhibited the mycelial growth of M.extelata by 7.6%,and the volatile product of P.trachicarpicola resulted in sparse mycelia and decreased aerial hyphae at 6 d.For the infected M.extelata ascocarps,the activity of SOD reached the highest at 2 d;the activity of POD was the highest at 1 d;and the activity of CAT was the lowest at 2 d.In the experimental range(72 h),M.extelata extract promoted the germination of WLM5 conidia.After infection,WLM5 produced cell wall degrading enzymes,and the activities of neutral protease,β-1,3 glucanase and chitinase were the highest at 15 d,6 d and 12 d,respectively.These results provided a reference for exploring the pathogenic mechanism of P.trachicarpicola.
