摘 要:目的探讨苍耳亭通过调节微小RNA(miR)-27a-3p靶向胸腺基质淋巴细胞生成素(TSLP)治疗过敏性鼻炎(AR)合并哮喘的作用机制。方法用卵清蛋白(OVA)致敏和鼻部滴注攻击法建立AR合并哮喘大鼠模型。将50只大鼠分为对照组(等体积0.9%NaCl)、模型组(构建AR模型合并哮喘+等体积0.9%NaCl)、实验组(构建AR模型合并哮喘+100mg·kg^(-1)苍耳亭)、miR-27a-3p inhibitor组(在实验组基础上尾静脉注射0.5nmol·μL^(-1)miR-27a-3p inhibitor10μL),si-TSLP组(在miR-27a-3p inhibitor组基础上尾静脉注射0.5nmol·μL^(-1)si-TSLP 10 μL),每组10只。用酶联免疫吸附试验法检测大鼠血清免疫球蛋白E(IgE)、白细胞介素-2(IL-2)、IL-13和肿瘤坏死因子-α(TNF-α)的水平,用黄嘌呤氧化酶法检测超氧化物歧化酶(SOD)水平,用硫代巴比妥酸法(TBA)检测丙二醛(MDA)水平。结果对照组、模型组、实验组、miR-27a-3p inhibitor组和si-TSLP组的IgE水平分别为(18.33±3.53)、(89.95±17.62)、(55.70±10.08)、(78.43±15.30)和(47.87±9.44)ng·mL^(-1),IL-2水平分别为(8.01±1.36)、(19.61±3.94)、(14.12±2.51)、(17.33±3.18)和(11.89±2.03)pg·mL^(-1),IL-13水平分别为(6.79±1.33)、(34.15±7.02)、(24.70±5.13)、(35.97±7.24)和(20.53±4.26)pg·mL^(-1),TNF-α水平分别为(94.08±19.07)、(312.47±58.61)、(209.78±41.49)、(296.42±55.99)和(187.45±37.28)pg·mL^(-1),SOD水平分别为(29.14±5.04)、(13.25±2.63)、(24.19±4.89)、(17.28±3.16)和(33.94±5.87)U·mg prot^(-1),MDA水平分别为(2.26±0.51)、(4.43±0.72)、(3.17±0.58)、(3.94±0.69)和(2.62±0.45)nmol·mg prot^(-1)。对照组的上述指标与模型组比较,模型组的上述指标与实验组比较,实验组的上述指标与miR-27a-3p inhibitor组比较,miR-27a-3p inhibitor组的上述指标与si-TSLP组比较,在统计学上差异均有统计学意义(P<0.05,P<0.01)。结论苍耳亭对大鼠的AR合并哮喘具有改善氧化应激、减轻炎症的治疗作用,其机制可能与调节miR-27a-3p靶向TSLP有�Objective To explore the mechanism of action of tanthine in the treatment of allergic rhinitis(AR)complicated with asthma in rats by regulating microRNA-27a-3p(miR-27a-3p)targetingthymic stromal lymphopoietin(TSLP).Methods The AR-asthma rat model was established using ovalbumin(OVA)sensitization and nasal drip attack method.Fifty rats were divided into control group(equal volume 0.9%NaCl),model group(AR-asthma model+equal volume 0.9%NaCl),experimental group(AR-asthmamodel+100mg·kg^(-1)xanthortin)and miR-27a-3p inhibitor group(caudal vein injection of 0.5 nmol·μL^(-1)miR-27a-3p inhibitor 10μL on the basis of the experimental group),si-TSLP group(0.5 nmol·μL^(-1)si-TSLP 10μL intravenously injected on the basis of miR-27a-3p inhibitor group),10 rats in each group.Serum immunoglobulin E(IgE),interleukin-2(IL-2),IL-13 and tumor necrosis factor-α(TNF-α)levels of rats were detected by enzyme-linked immunosorbent assay.The level of superoxide dismutase(SOD)was detected by xanthoxine oxidase method.The level of malonaldehyde(MDA)was detected by thiobarbituric acid method(TBA).Results The levels of IgE in control group,model group,experimental group,miR-27a-3p inhibitor group and si-TSLP group were(18.33±3.53),(89.95±17.62),(55.70±10.08),(78.43±15.30)and(47.87±9.44)ng·mL^(-1),respectively;IL-2 levels were(8.01±1.36),(19.61±3.94),(14.12±2.51),(17.33±3.18)and(11.89±2.03)pg·mL^(-1),respectively;IL-13 levels were(6.79±1.33),(34.15±7.02),(24.70±5.13),(35.97±7.24)and(20.53±4.26)pg·mL^(-1),respectively;TNF-αlevels were(94.08±19.07),(312.47±58.61),(209.78±41.49),(296.42±55.99)and(187.45±37.28)pg·mL^(-1),respectively;SOD levels were(29.14±5.04)(13.25±2.63),(24.19±4.89),(17.28±3.16)and(33.94±5.87)U·mg prot^(-1),respectively;MDA levels were(2.26±0.51),(4.43±0.72),(3.17±0.58),(3.94±0.69)and(2.62±0.45)nmol·m gprot^(-1),respectively.The above indicators were compared between control group and model group,model group and experimental group,experimental group and miR-27a-3p inhibitor group,miR
