摘  要:目的探讨超声靶向微泡爆破(UTMD)技术介导基质细胞衍生因子-1(SDF-1)转染增强三维(3D)培养骨髓间充质干细胞(BMSCs)心脏归巢效率。方法制备载SDF-1基因生物素化微泡(BMBs)(SDF-1/BMBs)。将24只SD大鼠分组:二维(2D)培养BMSCs、3D BMSCs、3D BMSCs+BMBs、3D BMSCs+SDF-1/BMBs。UTMD介导SDF-1基因于心脏释放。14 d后各组经尾静脉注射相应BMSCs。移植后48 h检测肺脏和心脏BMSCs数量。结果2D BMSCs组困于肺脏的数量(12.40±2.88)多于3D BMSCs;3D BMSCs+SDF-1/BMBs组心脏归巢数量最多(8.80±3.35)。结论3D BMSCs细胞直径小于2D BMSCs,通过肺循环较多;UTMD介导SDF-1基因转染可增加3D BMSCs心脏归巢数量。Objective To explore the efficiency of ultrasound-targeted microbubble destruction(UTMD)technique mediated stromal cell derived factor-1(SDF-1)transfection in cardiac homing of three-dimensional(3D)sphere cultured bone marrow mesenchymal stem cells(BMSCs).Methods Biotinylated microbubbles(BMBs)loaded with SDF-1 gene(SDF-1/BMBs)were prepared.Then 24 Sprague Dawley(SD)rats were divided into four groups:two-dimensional(2D)cultured BMSCs;3D BMSCs;3D BMSCs+BMBs;3D BMSCs+SDF-1/BMBs.UTMD-mediated SDF-1 gene local-delivery to the heart.Fourteen days later,each group was injected with BMSCs via tail vein respectively.Next the BMSCs in the lung and heart were measured 48 hours after transplantation.Results The quantity of 2D BMSCs trapped inside the lungs(12.40±2.88)was significantly greater than that of 3D BMSCs.Notably,the amount of BMSCs within the 3D BMSCs+SDF-1/BMBs group exhibited the highest value(8.80±3.35)homing to the heart.Conclusions The diameter of 3D BMSCs is smaller than that of 2D BMSCs,which were trapped in the lungs less.Furthermore,SDF-1 gene transfection mediated by UTMD can significantly augment the homing capacity of 3D BMSCs.
