摘  要:目的探讨厚胞汤调节成纤维细胞对防治宫腔粘连的作用机理。方法将10只健康SD大鼠随机数字表法分成空白对照组和厚胞汤组,空白对照组灌胃等量生理盐水,厚胞汤组灌胃厚胞汤2 m L/只·次,每天2次以制备空白血清及含药血清,将经转化生长因子-β1(TGF-β1)刺激后的成纤维细胞分为空白对照组和厚胞汤组,采用CCK8法检测两组细胞增殖情况,流式细胞法检测细胞凋亡水平、细胞周期变化,Western blot检测两组细胞的TGF-β1、信号转导蛋白家族(Smads)表达水平。结果与空白对照组比较,厚胞汤组的细胞增殖率显著降低[第2天:(1.05±0.04)%比(1.19±0.02)%,P<0.01;第3天:(1.11±0.02)%比(1.36±0.02)%,P<0.01;第4天:(1.18±0.02)%比(1.55±0.02)%,P<0.001;第5天:(1.16±0.02)%比(1.89±0.03)%,P<0.01];细胞凋亡率显著升高[(10.19±0.72)%比(6.31±0.17)%,P<0.01];细胞周期G1期比例降低[(48.78±1.51)%比(52.30±0.56)%,P<0.05],G2期比例升高[(27.56±0.64)%比(22.86±0.88)%,P<0.01]。Western blot结果显示,厚胞汤组TGF-β1、Smad3表达显著下调。结论厚胞汤可能通过TGF-β/Smads通路有效抑制成纤维细胞纤维化表达,从而对宫腔粘连起到防治作用。Objective To investigate the mechanism of Houbao Decoction in the prevention and treatment of intrauterine adhesions by regulating fibroblast.Methods Ten healthy SD rats were randomly divided into a control group and a Houbao Decoction group using a random number table.The Houbao Decoction group was gavaged with 2 mL per rat per dose of Houbao Decoction,twice a day,while the control group was gavaged with an equal amount of physiological saline.Blank serum and drug-containing serum were collected.The fibroblast stimulated by transforming growth factor-β1(TGF-β1)were divided into a control group and a Houbao Decoction group.The cell proliferation in both groups was detected using the CCK8 method.Apoptosis and cell cycle were analyzed using flow cytometry.The expression levels of TGF-β1 and the signal transduction protein family(Smads)in both groups were detected using western blotting.Results Compared with the control group,the cell proliferation rate in the Houbao Decoction group was significantly reduced[Day 2:(1.19±0.02)vs.(1.05±0.04)%,P<0.01;Day 3:(1.36±0.02)vs.(1.11±0.02)%,P<0.001;Day 4:(1.55±0.02)vs.(1.18±0.02)%,P<0.001;Day 5:(1.89±0.03)vs.(1.16±0.02)%,P<0.001].The cell apoptosis rate was significantly increased[(10.19±0.72)vs.(6.31±0.17)%,P<0.01].The proportion of cells in the G1 phase of the cell cycle was decreased[(48.78±1.51)vs.(52.30±0.56)%,P<0.05],while the proportion of cells in the G2 phase was increased[(27.56±0.64)vs.(22.86±0.88)%,P<0.01].Conclusion Houbao Decoction may inhibit the fibrotic expression of fibroblast by the TGF-β/Smads pathway,thereby playing a role in the prevention and treatment of intrauterine adhesions.
