基于网络药理学探索抑酸护胃方治疗胃食管反流病的作用机制

摘　　要：目的 分析抑酸护胃方治疗胃食管反流病(GERD)的作用机制。方法 基于网络药理学,通过在线数据库获得抑酸护胃方的活性成分和靶点以及GERD的靶点,将二者的交叉靶点作为潜在靶点,通过STRING数据库构建蛋白互作关系,通过Metascape数据库进行基因本体论(GO)及京都基因与基因组百科全书(KEGG)分析。结果 抑酸护胃方共有429个活性成分和2005个靶点,GERD有949个靶点,潜在靶点320个;通过GO分析得到生物过程3049个、细胞组分183个、分子功能298个;通过KEGG富集分析得到223条信号通路。结论 抑酸护胃方可能是通过丁香酚、肉桂醛等活性成分,作用于β-肌动蛋白(ACTB)、蛋白激酶B(AKT1)等多个靶点,参与多条信号通路,通过抑菌消炎、抗氧化、保护胃黏膜、抗癌等药理作用治疗GERD,为临床应用抑酸护胃方治疗GERD提供了一定的数据支撑,同时也为治疗GERD的新药开发提供了一些参考。Objective To analysis the mechanism of the treatment of gastroesophageal reflux disease(GERD)with Yisuan Huwei pre-scription.Methods Based on network pharmacology,the active components and targets of Yisuan Huwei prescription,as well as the targets of GERD,were obtained through online databases.The cross targets of the two were used as potential targets.The STRING da-tabase was used to construct protein interaction relationships,and the Metascape database was used for GO and KEGG analysis.Re-sults There were 429 active ingredients and 2005 targets in Yisuan Huwei prescription,949 targets in GERD,and 320 potential tar-gets.GO analysis obtained 3049 biological processes,183 cell components,and 298 molecular functions.KEGG enrichment analysis revealed 223 signal pathways.Conclusion Yisuan Huwei prescription may act on multiple targets such as ACTB and AKT1 etc.through active ingredients such as eugenol and cinnamaldehyde etc.,participate in multiple signal pathways,finally treat GERD through pharmacological actions such as bacteriostasis and anti-inflammatory,antioxidant,protection of gastric mucosa,and antican-cer etc.This provides certain data support for clinical application of Yisuan Huwei prescription in treating GERD,and also provides some reference for the development of new drugs for treating GERD.
