摘 要:目的 探讨刺老苞防治骨质疏松症(OP)的分子机制。方法 将36只SD大鼠随机分为空白组,模型组,阳性对照组(维生素D组),刺老苞水提取物高、中、低剂量组,各6只。以苏木素-伊红(HE)染色观察大鼠股骨病理学变化,逆转录-聚合酶链反应(RT-PCR)法检测股骨组织细胞色素P450羟化酶(CYP17A1)、细胞色素P450 19A1(CYP19A1)mRNA表达情况。结果 与空白组比较,模型组大鼠HE染色镜下骨小梁明显变细伴断裂、稀疏,骨小梁数量重度减少、间隙增大。与模型组比较,刺老苞水提取物高剂量组大鼠可见骨小梁数量明显增多、骨小梁局部明显增粗,且刺老苞水提取物高剂量组优于刺老苞水提取物低、中剂量组。与空白组比较,模型组大鼠股骨中CYP17A1、CYP19A1 mRNA相对表达量显著降低(P<0.01);与模型组比较,刺老苞水提取物高剂量组、阳性对照组大鼠股骨中CYP17A1、CYP19A1 mRNA相对表达量显著升高(P<0.01)。结论 刺老苞通过调节股骨组织CYP17A1、CYP19A1 mRNA表达情况,发挥干预OP的作用。Objective To explore the molecular mechanism of Aralia Chinensis L in preventing osteoporosis(OP).Methods A total of 36 SD rats were randomly divided into blank group,model group,positive control group(vitamin D group),high-dose,medium-dose and low-dose groups with 6 rats each.The pathological changes of femur were observed by hematoxylin-eosin(HE)staining,and the mRNA expressions of cytochrome P450 hydroxylase(CYP17A1)and cytochrome P45019A1(CYP19A1)were detected by reverse transcription-polymerase chain reaction(RT-PCR).Results Compared with the blank group,the trabecular thinning with fracture,sparsity,severely reduced trabecular number and increased gap.Compared with the model group,the number of trabecules increased significantly in the increasing dose of the group,and the local thickening of the trabecules increased significantly,while the increased dose of the group was better than the low and medium dose groups.Compared with the blank group,the relative expression of CYP17A1 and CYP19A1 mRNA in the model group(P<0.01).Compared with the model group,the relative expression of CYP17A1 and CYP19A1 mRNA in the group and the positive control group(P<0.01).Conclusion By regulating the expression of CYP17A1 and CYP19A1mRNA in femoral tissue,it plays its role in intervening in OP.
