摘 要:目的探讨中脑腹侧被盖核(NAc)中γ-氨基丁酸B型受体(GABABR)对可卡因成瘾大鼠的戒断症状及认知功能的影响。方法建立可卡因依赖动物模型,将SD大鼠随机分为正常对照组、生理盐水组和GABABR激动剂巴氯芬(BLF)组,每组20只,应用条件性位置偏好(CPP)实验来评价大鼠对可卡因的依赖程度,通过IntelliCage全自动监测系统来观察NAc中GABABR对可卡因成瘾大鼠戒断症状和认知功能的影响。结果CPP实验结果表明,3组大鼠给予可卡因干预后,在伴药箱的停留时间明显增长,出入次数明显增加。鼻触学习中,BLF组大鼠在给药2 h内的饮水次数明显多于正常对照组和生理盐水组(P<0.05);空间探索测试中,BLF组大鼠访问先前开放角落的次数明显多于其他2组(P<0.05),正常对照组大鼠和生理盐水组大鼠对比无明显变化(P>0.05)。结论NAc中GA BABR参与可卡因成瘾的调节,其激动剂BLF可使依赖可卡因的大鼠出现戒断症状,改善其认知功能。Objective To investigate the effect of GABABR in nucleus accumbens on withdrawal symptoms and cognitive function in cocaine-addicted rats.Methods Cocaine dependence rat model was established,and rats were randomly divided into normal control group(n=20),the saline group(n=20)and the BLF group(n=20).The degree of cocaine dependence in rats were evaluated by conditioned place preference experiments.The effects of GABABR in nucleus accumbens on withdrawal symptoms and cognitive function in cocaine-addicted rats by Intellicage automatic monitoring system were observed.Results After cocaine intervention,the residence time and the number of entry and exit in the three groups of rats increased significantly in the companion box.Nose-touch learning:the frequency of water consumed in the BLF group was significantly higher than that in the normal control group and the saline group within two hours of administration(P<0.05).Space exploration test:the number of visits to the previously opened corners by rats in the BLF group was higher than that in normal control group within two hours after administration(P<0.05),but there was no significant change between the normal control group and the normal saline group(P>0.05).Conclusion GABABR in nucleus accumbens is involved in the regulation of cocaine addiction,and its agonist BLF can cause withdrawal symptoms and improve cognitive function in cocaine-dependent rats.
