摘　　要：目的 探讨2型糖尿病(T2DM)大鼠回肠组织铁死亡对胰高血糖素样肽-1(GLP-1)的影响及玉液汤的作用。方法 采用高脂饮食联合腹腔注射链脲佐菌素(STZ)制备T2DM大鼠模型,随机分为模型组、高剂量组和低剂量组,每组8只。另选取8只同龄大鼠作为对照组。采用连续灌胃给药4周,进行口服糖耐量实验(OGTT),并计算血糖曲线下面积(AUC)。检测血空腹胰岛素(FINS),计算胰岛素分泌指数(HOMA-β)及胰岛素抵抗指数(HOMA-IR)。检测回肠组织Fe^(2+)、丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶4(GPX4)、铁死亡抑制蛋白1(FSP1)、胰高血糖素样肽-1(GLP-1)水平。结果 与模型组比较,治疗后高剂量组及低剂量组血糖、AUC、HOMA-IR、回肠组织中Fe^(2+)和MDA显著下降(P<0.05),FINS、HOMA-β、回肠组织中SOD、GPX4、FSP1及GLP-1显著上升(P<0.05)。结论 玉液汤能改善T2DM大鼠回肠组织铁死亡,上调GLP-1,减轻胰岛素抵抗。Objective To observe the effect of Yuye Decoction on improving GLP-1 in type 2 diabetes mellitus(T2DM)rats by regulating ferroptosis in ileum tissue.Methods T2DM rat model was prepared by high fat diet combined with intraperitoneal injection of streptozotocin(STZ).Rats were randomly assigned into model group,high dose group and low dose group,with 8 rats in each group.Eight age-matched rats were included in the control group.After 4 weeks of continuous intragastric administration,oral glucose tolerance tests(OGTT)were carried out,and the area under the curve(AUC)was calculated.Fasting insulin(FINS)was measured.Homeostatic model assessment forβ-cell function(HOMA-β),homeostasis model assessment for insulin resistance(HOMA-IR)were calculated.Fe^(2+),malondialdehyde(MDA),superoxide dismutase(SOD),glutathione peroxidase 4(GPX4),ferroptosis suppressor protein 1(FSP1)and glucagon-like peptide-1(GLP-1)in rat ileum tissues was detected.Results Compared with those of the model group,rats in the high dose group and low dose group had significantly decreased blood glucose level,AUC,HOMA-IR,Fe^(2+)and MDA(P<0.05),but increased FINS,HOMA-β,SOD,GPX4,FSP1 and GLP-1levels(P<0.05).Conclusion Yuye Decoction could ameliorate ferroptosis in ileum tissue of T2DM rats,upregulate GLP-1 and reduce insulin resistance.
