摘 要:目的:建立安全可靠的放射性皮炎小鼠模型,观察中药制剂复方紫草油的治疗效果,并研究对其水通道蛋白3(AQP3)和丝聚蛋白(FLG)表达的影响,为拓展及开发放射性皮炎的中药制剂提供依据。方法:将放射性皮炎小鼠随机分为健康对照组、模型组(单纯辐照组)、复方紫草油组、透明质酸组和卤米松软膏组,造模后治疗组分别用复方紫草油、透明质酸和卤米松软膏外用于小鼠皮损处,每日2次,共18日,对治疗前后放射性皮炎分级评分、皮损组织病理、免疫组化,表皮层及真皮层的厚度、皮损中CD3^(+)T淋巴细胞、CD11c^(+)树突细胞、CD68^(+)巨噬细胞以及AQP3、FLG的表达水平进行对照。结果:与单纯辐照组比较,复方紫草油、透明质酸、卤米松软膏组治疗小鼠放射性皮炎分级评分具有统计学意义(P<0.05),表皮层及真皮层厚度显著降低(P<0.05),组织病理显示治疗后皮损中炎性细胞浸润减轻,免疫组化显示皮损中CD3^(+)T淋巴细胞、CD11c^(+)树突细胞、CD68^(+)巨噬细胞浸润情况均有缓解,AQP3与FLG的表达有所恢复。结论:复方紫草油能缓解小鼠放射性皮炎,减轻表皮和真皮水肿,降低皮损中炎细胞浸润,并能调控皮损中FLG及AQP3的表达,治疗效果与透明质酸和卤米松软膏相当。Objective:To establish a safe and reliable mouse model of radiation dermatitis,to observe the therapeutic effect of Compound Radix Arnebiae Oil on X-ray-induced radiation dermatitis,and to explore their regulatory effect on filaggrin(FLG)and aquaporin 3(AQP3).This will provide a basis for the development of traditional Chinese medicine preparations for radiation dermatitis.Methods:Mice with radiation dermatitis were randomly divided into healthy control group,model group(simple irradiation group),Compound Radix Arnebiae Oil group,hyaluronic acid group and halometasone ointment group.After modeling,the treatment group applied Compound Radix Arnebiae Oil,hyaluronic acid and halometasone ointment to the skin lesions of mice,twice a day for 18 days.Before and after treatment,the grading score of radiation dermatitis,histopathology and immunohistochemistry of the skin lesions,the thickness of epidermis and dermis,CD3^(+)T lymphocytes,CD11c^(+)dendritic cells,CD68^(+)macrophages,AQP3 and FLL in the skin lesions were evaluated.Results:Compared with the simple irradiation group,the grading scores of radiation dermatitis in mice treated with Compound Radix Arnebiae Oil,hyaluronic acid group and halometasone Ointment were statistically significant(P<0.05),and the thickness of epidermis and dermis decreased significantly(P<0.05).Histopathology showed that the infiltration of inflammatory cells in skin lesions was reduced after treatment.Immunohistochemistry showed that the infiltration of CD3^(+)T lymphocytes,CD11c^(+)dendritic cells and CD68^(+)macrophages in the lesions was alleviated,and the expressions of AQP3 and FLG were restored.Conclusion:Compound Radix Arnebiae Oil can relieve radiation dermatitis in mice,reduce edema of epidermis and dermis,reduce infiltration of inflammatory cells in skin lesions,and regulate the expression of FLG and AQP3 in skin lesions.The therapeutic effect is equivalent to that of hyaluronic acid group and halometasone ointment.
