摘　　要：目的建立应用高效液相色谱法(HPLC)同时测定尿感安丸中大车前苷、甘草苷、芦荟大黄素、大黄素、大黄酚和大黄素甲醚含量的方法。方法采用HPLC测定样品含量。采用中谱红RD-C18(4.6 mm×250.0 mm,5.0μm)色谱柱,以乙腈-0.1%甲酸溶液为流动相,梯度洗脱,检测波长284 nm,流速1.0 mL/min,柱温35℃。结果大车前苷、甘草苷、芦荟大黄素、大黄素、大黄酚、大黄素甲醚的线性范围分别为18.58~371.62 ng(r=0.9990)、5.09~101.85 ng(r=0.9999)、1.56~31.32 ng(r=0.9995)、1.71~34.17 ng(r=0.9999)、15.39~307.92 ng(r=0.9998)和4.16~83.32 ng(r=0.9990);精密度、稳定性和重复性试验的相对标准偏差(RSD)均<1.40%(n=6);平均加样回收率为96.42%~104.50%,RSD为0.41%~1.13%(n=6)。结论HPLC准确简便,专属性好,精密度、稳定性、重复性和加样回收率均符合方法学验证要求,可以为尿感安丸的有效成分研究及质量标准制定提供参考。Objective To establish a method for the simultaneous determination of the components of plantamajoside,liquiritin,aloe-emodin,emodin,chrysophanol and physcion in Niaogan'an Pills by high performance liquid chromatography(HPLC).Methods The sample components were determined by HPLC.A Zhongpu Hong RD-C18(4.6 mm×250.0 mm,5.0μm)chromatographic column was used,and the mobile phase was composed of acetonitrile-0.1%formic acid solution by gradient elution.The detection wavelength was 284 nm,the flow rate was 1.0 mL/min,and the column temperature was 35℃.Results The linear ranges of plantamajoside,liquiritin,aloe-emodin,emodin,chrysophanol and physcion were 18.58-371.62 ng(r=0.9990),5.09-101.85 ng(r=0.9999),1.56-31.32 ng(r=0.9995),1.71-34.17 ng(r=0.9999),15.39-307.92 ng(r=0.9998)and 4.16-83.32 ng(r=0.9990),respectively.The relative standard deviations(RSD)of precision,stability and repeatability tests were all less than 1.40%(n=6).The average recovery rate was 96.42%-104.50%,and the RSD was 0.41%-1.13%(n=6).Conclusions HPLC is accurate and simple with good specificity,and it meets the requirements of methodology validation in terms of precision,stability,repeatability,and recovery rate,which can provide a reference for the research of effective ingredients and the formulation of quality standards for Niaogan'an Pills.
