健脾益气利水汤缓解顺铂所致小鼠肾毒性的研究

摘  要:目的:探讨健脾益气利水汤缓解顺铂所致小鼠肾毒性的作用。方法:选取40只BALB/c小鼠随机分为对照组、模型组、预防组及解毒组。通过静脉注射顺铂构建肾损伤模型并灌胃健脾益气利水汤,检测血清中肾功能指标、肾组织氧化应激指标及炎性指标、PAS染色分析肾组织病理特点。结果:与模型组对比,预防组及解毒组尿NAG、血清Scr及BUN水平均低于模型组(P<0.05),相较于模型组,预防组和解毒组肾组织中MDA降低,而SOD及GSH水平明显升高(P<0.05),预防组Pt蓄积量明显减低(P<0.05);与预防组比较,解毒组SOD明显偏低,而MDA及Pt蓄积量则偏高(P<0.05)。与模型组相比,预防组与解毒组小鼠肾组织KIM mRNA表达量明显下降,且预防组KIM mRNA表达量低于解毒组(P<0.05)。结论:健脾益气利水汤能够改善顺铂所致肾毒性小鼠的肾脏功能。相较于解毒组,预防组给药能显著抑制顺铂引发氧化应激反应,下调KIM-1表达水平从而更好保护肾脏功能。Objective:To explore the effect of Jianpi Yiqi Lishui Tang on alleviating nephrotoxicity induced by cisplatinum in mice.Method:Forty BALB/c mice were randomly divided into a control group,a model group,a prevention group,and a detoxification group.Constructing a kidney injury model by intravenous injection of Shunlead and administering Jianpi Yiqi Lishui decoction by gavage,detecting renal function indicators,renal tissue oxidative stress indicators,and inflammatory indicators in serum;PAS staining analysis of renal tissue pathological characteristics.Result:Compared with the model group,the urine NAG,serum Scr,and BUN levels in the prevention and detoxification groups were lower than those in the model group(P<0.05).Compared with the model group,the MDA levels in the kidney tissue of the prevention and detoxification groups decreased,while the levels of SOD and GSH increased significantly(P<0.05).The accumulation of Pt in the prevention group decreased significantly(P<0.05);Compared with the prevention group,the detoxification group had significantly lower SOD levels,while MDA and Pt accumulation levels were higher(P<0.05).Compared with the model group,the expression levels of KIM mRNA in the kidney tissues of mice in the prevention group and detoxification group were significantly decreased,and the expression level of KIM mRNA in the prevention group was lower than that in the detoxification group(P<0.05).Conclusion:Jianpi Yiqi Lishui Tang can improve renal function in cisplatin induced nephrotoxicity mice.Compared with the detoxifi.
