摘  要:逆转录病毒及其载体的逆转录特性和遗传稳定性使其被广泛应用于各个领域。逆转录病毒(如小鼠白血病病毒)可作为指示病毒应用于生物制品生产工艺中的病毒清除、灭活验证领域。逆转录病毒载体通过基因传递技术靶向特定组织或细胞进行长期转基因表达,广泛应用于基因治疗、基因改造等领域。随着逆转录病毒及其载体的使用范围扩大,生产高滴度病毒或病毒载体的能力对于避免增加生产成本及减少下游纯化压力非常重要。如何优化病毒滴度是病毒清除、灭活验证或基因传递技术领域亟待解决的问题。本文综述了逆转录病毒及其载体滴度的优化进展和策略,总结了影响逆转录病毒滴度的潜在因素,以期为未来逆转录病毒滴度的优化提供参考。Retroviruses and retroviral vectors are used widely in various fields due to their reverse-transcription properties and genetic stability.Certain retroviruses,such as mouse leukemia virus,can be used as“model”viruses in viral clearance/inactivation validation during biologics manufacture.Tissue-specific or cell-specific targeting of retroviral vectors has been established through gene-delivery technology for long-term transgenic expression,and is used widely in gene therapy,gene modification,and other fields.As the use of retroviruses and their vectors expands,the ability to produce high-titer viruses or viral vectors reliably is important to avoid increasing production costs and reduce downstream purification stress.How to optimize viral titers is an urgent problem in viral clearance/inactivation verification or gene-delivery technology.Herein,progress and strategies in the development and optimization of retroviruses and their vector titers are reviewed.The factors that may affect retroviral titers are summarized to provide a reference for future optimization of retrovirus titers.
