摘 要:目的 探究舒芬太尼(SUF)对白细胞介素-1β(IL-1β)诱导的软骨细胞损伤的作用及其与酪氨酸激酶2(JAK2)/信号传导与转录激活子3(STAT3)信号通路的调节关系。方法 将大鼠软骨细胞随机分为对照组(NC组)、IL-1β组、SUF低浓度(SUF-L)+IL-1β组(SUF-L+IL-1β组)、SUF高浓度(SUF-H)+IL-1β组(SUF-H+IL-1β组)、SUF-H+IL-1β+STAT3激活剂Colivelin组(SUF-H+IL-1β+Colivelin组);CCK-8法和克隆形成实验检测细胞增殖情况,流式细胞术检测细胞凋亡情况;qRTPCR测定JAK2和STAT3的mRNA水平;Western blot检测JAK2/STAT3信号通路相关蛋白表达;酶联免疫吸附法测定各组细胞培养液IFN-γ、IL-6和IL-8表达水平。结果 与NC组相比,IL-1β组细胞存活率及克隆形成数量降低,细胞凋亡率、JAK2和STAT3 mRNA相对表达量、p-JAK2/JAK2和p-STAT3/STAT3值以及细胞培养液IFN-γ、IL-6和IL-8表达水平均升高(P<0.05);与IL-1β组相比,SUF-L+IL-1β组、SUF-H+IL-1β组细胞对应指标变化趋势与上述相反(P<0.05);Colivelin逆转了高剂量SUF对IL-1β诱导的软骨细胞损伤的保护作用(P<0.05)。结论 SUF能够通过下调JAK2/STAT3信号通路对IL-1β诱导的软骨细胞损伤起保护作用。Objective To investigate the effect of sufentanil(SUF)on interleukin-1β(IL-1β)-induced chondrocyte damage and its regulatory relationship on the Janus activated kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3)signaling pathway.Methods Rat chondrocytes were randomly separated into control group(NC group),IL-1βgroup,SUF low concentration(SUF-L)+IL-1βgroup(SUF-L+IL-1βgroup),SUF high concentration(SUF-H)+IL-1βgroup(SUF-H+IL-1βgroup),and SUF-H+IL-1β+STAT3 activator Colivelin group(SUF-H+IL-1β+Colivelin group).CCK-8 method and clonal formation assay were applied to detect the proliferation of chondrocytes,while flow cytometry was used to detect the apoptosis;qRT-PCR method was applied to detect the relative mRNA expression levels of JAK2 and STAT3;Western blot was used to detect the expression level of JAK2/STAT3 signaling pathway related proteins;ELISA was applied to determine the expression levels of IFN-γ,IL-6 and IL-8 in cell culture media of each group.Results Compared with the NC group,the cell survival rate and the clones formation number of the IL-1βgroup decreased,while the apoptosis rate,relative expression levels of JAK2 and STAT3 mRNA,the values of p-JAK2/JAK2 and p-STAT3/STAT3,and the expression levels of IFN-γ,IL-6 and IL-8 in the cell culture medium all increased(P<0.05);compared with the IL-1βgroup,the changes in corresponding indicators of chondrocytes in the SUF-L+IL-1βgroup and SUF-H+IL-1βgroup were opposite to the above(P<0.05);Colivelin reversed the protective effects of high-dose SUF on IL-1β-induced chondrocyte damage(P<0.05).Conclusion SUF can protect chondrocytes from IL-1β-induced damage by down-regulating the JAK2/STAT3 signaling pathway.
