摘 要:为了研究贡柑和沃柑无籽形成的分子机理,选取了贡柑和沃柑作为研究对象,成功克隆了它们的E3泛素连接酶U-box基因。通过测序结果分析,发现贡柑和沃柑的U-box基因存在15个核苷酸差异,导致贡柑中有4个氨基酸发生了改变。通过序列比对,发现沃柑和无籽沙糖橘的E3泛素连接酶U-box基因相似度为98.19%,贡柑和无籽沙糖橘的E3泛素连接酶U-box基因相似度为97.72%,而沃柑和贡柑的E3泛素连接酶U-box基因相似度高达99.47%。In order to study the molecular mechanism of seedless formation in Citrus reticulata cv.Gonggan and Orah mandarin,Citrus reticulata cv.Gonggan and Orah mandarin were selected as research objects,and their E3 ubiquitin ligase U-box genes were successfully cloned.After analyzing the sequencing results,it was found that there were 15 nucleotide differences in the U-box genes of Citrus reticulata cv.Gonggan and Orah mandarin,resulting in changes in 4 amino acids in Citrus reticulata cv.Gonggan.Through sequence alignment,it was found that the similarity of E3 ubiquitin ligase U-box genes between Orah mandarin and Citrus reticulata‘Wuzishatangju’was 98.19%,while the similarity of E3 ubiquitin ligase U-box genes between Citrus reticulata cv.Gonggan and Citrus reticulata‘Wuzishatangju’was 97.72%.The similarity of E3 ubiquitin ligase U-box genes between Orah mandarin and Citrus reticulata cv.Gonggan was as high as 99.47%.
