摘 要:[目的]进一步探索青牛胆人工扩繁育苗技术,有效促进野生青牛胆资源的保护和可持续开发利用。[方法]以雷公山自然保护区野外采集的青牛胆种子及藤蔓为试验材料,采用单因素试验设计,研究不同基质类型对种子萌发的影响与不同基质类型、不同生长激素及浓度对扦插繁殖的影响。[结果]播种基质为腐殖土+河沙(1∶1)时种子发芽率最高,达(88.33±1.53)%;扦插基质为壤土+河沙(1∶1)时,扦插成活率最高,达(54.45±6.49)%;IAA溶液处理浓度为1.0 mg/L时,扦插成活率最高,达(56.67±5.77)%,NAA溶液处理浓度为1.0 mg/L时,扦插成活率最高,达(75.56±8.39)%,ABT溶液处理浓度为1.0mg/L时,扦插成活率最高,达(68.89±5.09)%。[结论]播种基质以腐殖土+河沙(1∶1)青牛胆种子萌发效果最好,可作为青牛胆播种育苗基质使用;扦插基质以壤土+河沙(1∶1)扦插效果最优;从3种不同生长激素处理扦插的成活率来看,均提高了青牛胆扦插成活率,以1.0 mg/L NAA溶液处理扦插成活率最高,达(75.56±8.39)%,综合扦插效果最优。[Objective]In order to further explore the artificial propagation and seedling cultivation technology of Tinospora sagitata,and effectively promote the protection and sustainable development and utilization of wild Tinospora sagitata resources.[Method]Using Tinospora sagittata seeds and vines collected in the Leigongshan Nature Reserve as experimental materials,a single factor experimental design was used to study the effects of different substrate types on seed germination and the effects of different substrate types,growth hormones,and concentrations on cutting propagation.[Result]The results showed that the highest seed germination rate(88.33±1.53)%was achieved when the sowing substrate was humus soil+river sand(1∶1);the highest survival rate(54.45±6.49)%was achieved when the cutting substrate was loam soil+river sand(1∶1);the highest survival rate(56.67±5.77)%was observed in cuttings treated with IAA solution at a concentration of 1.0 mg/L,the highest survival rate(75.56±8.39)%was observed in cuttings treated with NAA solution at a concentration of 1.0 mg/L,and the highest survival rate(68.89±5.09)%was observed in cuttings treated with ABT solution at a concentration of 1.0 mg/L.[Conclusion]Based on the above,the best germination effect of Tinospora sagitta seeds can be achieved by using humus soil+river sand(1∶1)as the sowing substrate,which can be used as the substrate for Tinospora sagitta seeding and seedling cultivation;the optimal cutting effect is achieved with a mixture of loam soil and river sand(1∶1)as the cutting substrate;from the perspective of three different growth hormone treatments,the survival rate of Tinospora sagittata cuttings was improved to varying degrees.The highest survival rate(75.56±8.39)%was observed in the NAA solution at a concentration of 1.0 mg/L,indicating the best overall cutting effect.
